Background The colon cancer, a familiar malignant tumor harmed human health, exhibited a high death rate and an increasing trend for incidence rate. The chief reason for causing death of cancer was malignant invade and metastasis by lymphatic. The research displayed that there were a lot of abnormal cytokine expression and heredity instability mutation of anti-oncogene during cancer occurrence and lymphatic metastasis. Intercellular adhesion molecule-1 (ICAM-1), an important transmembrane glycoprotein widely expressed on the surface of various normal cell, negatively expressed on endothelial cells of lymphatics but positively expressed on endothelial cells of blood vessel in normal condition. As linking specific ligand, lymohocyte function associated antigen 1 (LFA-1) existed on the surface of lymphocyte, ICAM-1 could induce lymphocyte attach to endothelial cells of blood vessel and traverse blood vessel, aggregate to antigen and took part in immune and inflammatory reaction. Vasse's study displayed that there were high expression of lymphocyte function associated antigen 1 (LFA-1), the ligand of ICAM-1, in cancer cells. There were few report in domestic and international research whether the endothelial cells of lymphatics expressed ICAM-1 or not in cancer tissue and whether ICAM-1/LFA-1 took part in the process of cancer cells attached to endothelial cells of lymphatics and metastasis. Dendritic cells is the important antigen presentation cells in body which can present tumor antigen to nature killed cells and cytotoxic cells, so can exercise the function of killing tumor. The research displayed that DCs was always the controlcenter in the body anti-tumor immune and The variation of the numbers of DCs in the tumor region can reflect the variation of the ability to anti-tumor associated with the tumor progression. To increase the numbers of DCs in tumor can obviously enhance the patients prognosis. The auto-transfuse of sensitivity DCs was already the hot spot for tumor therapy. It's important value to evaluate the ability of anti-tumor and prediction the patients prognosis and adoption the operative immune therapy for anti-tumor by research the numbers and distribution of DCs in tumor as the occurrence and lymphatic metastasis of colon cancer.For its production nm23Hi protein/NDPK could specifically suppress tumor metastasis, The nm23Hi gene, an important anti-oncogene located at 17 th chromosome (17q21), was named as anti-metastasis gene and was carried a lot of research which mostly concentrated on immunohistochemistry research of nm23Hi protein/NDPK expression. But there were few researches that related to heredity mutation of anti-metastasis gene nm23Hl. Objective (1) To study the expression of intercellular adhesion molecule-1 (ICAM-1) on lymphatics endothelial cells and the distribution of dendritic cells (DCs) as lymphatic metastasis of colon cancer. (2) To study the microsatellite instability (MSI) and the loss of heterozygosity (LOH) of D17S396 loci in Chinese 17th chromosomes influenced the expression of nm23Hi protein. (3) To clarify the relationship between the heredity instability of nm23Hi gene, the abnormal expression of adhesion molecule and the development of colon cancer, and to provide the experimental basis for clinical treatment.Methods (1) The technology of extraction DNA from paraffin-embedded material. (2) Polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and displaying band by ordinary silver stain. (3) Envision immunohistochemistry. (4) Leica-Qwin computer imaging techniques. (5) SPSS statistic software analyzed the experimental results. Results (1) ICAM-1 expression of lymphatic endothelium was negative innormal colon and positive in conlon cancer respectively. (2) The number of ICAM-4 positive lymphatics was gradually increased with depth of cancer invasion ( P<0.01).
