| Objective To explore the expression of IKKα, IκBα, NF-κB P65 of nuclear transcription factor NF-κB signal transduction pathway in laryngeal carcinoma patients.Methods The expression of IKKα, IκBα, NF-κB P65 was examined by immunohistochemical method in laryngeal carcinoma tissue of 40 cases and normal laryngeal tissue of 20 cases; The expression of IKKα, IκBα, NF-κB P65 was examined by western blot and the NF-κB DNA binding activity was examined by electrophoretic mobility shift assay(EMSA) in laryngeal carcinoma tissue of 20 cases and normal laryngeal tissues of 10 cases.Results The expression of IκB, NF-κB P65, and the NF-κB DNA binding activity in laryngeal carcinoma tissue was higher comparerd with normal laryngeal tissue, the expression of IκBαwas lower(P<0.05). The expression of Ikkα, NF-κB P65 in the carcinoma tissue with neck lymphnode metastasis was higher than that without neck lymph node metastasis, the expresion of IκBα was lower(P<0.05).Conclusion The expression of NF-κB signal transduction pathway is anomaly in the laryngeal carcinoma tissue and has closely relation with the occurrence, development and metastasis of laryngeal carcinoma. The inhibition of NF-κB signal transduction pathway can offer a new target of the treatment in laryngeal carcinoma. |
