Objective MMPs are a gene family of Zn2+ endopeptidases that digest various extracellular matrix (ECM) macromolecules. The activation and expression of MMP are related with many physiologic pathologic symptoms, such as inflammation, tissue fiber-ization and vasifaction, especially the nephrogenesis and metastasis of tumors, TIMPs are a gene family of multifunction factor, TIMP has an important role in the rebuilding of ECM and all kinds of pathologic process by suppressing the activation of MMP. We studied the expression of MMP-7 and TIMP-1 genes in 30 samples of human gastric carcinomas and the association, trying to find their effect on the nephrogenesis and development of gastric carcinoma and lay the theoretical and experimental foundation for gastric carcinoma therapy.Methods The expression of MMP-7 and TIMP-1 genes in 30 gastric carcinomas specimens and 30 corresponding adjacent normal gastric tissue specimens were detected by a semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR).Results 23(76. 7%) of the 30 gastric carcinomas specimens expressed MMP-7 mRNA, There was no expression of MMP-7 gene in the adjacent tissue of gastric carcinoma. There was highly significant statistic difference in the expression of MMP-7 gene between the gastric carcinoma and the adjacent tissue of gastric carcinoma (%2=37. 297, P<0. 001). 16(53. 3%) of the 30 gastric carcinomas specimens expressed TIMP-1 mRNA, 14(46. 7%) of the 30 adjacent tissue of gastric carcinomas specimens expressed TIMP-1 mRNA, there was no significant difference in the expression of TIMP-1 gene between the gastric carcinoma and the adjacent tissue of gastric carcinoma (x2=0.267, P>0. 05). The expression of MMP-7 and TIMP-1 gene in the 30 gastric carcinoma was not associated with the pathological differentiation or clinical stages of the gastric carcinomas(P>0. 05). 4The expressive quantity of MMP-7 gene in the low differentiated adenocarcinoma tissue and the gastric carcinomas tissue with lymph node metastasis was 2. 10 + 0. 77(x + s) and 1. 79 + 0. 88(5T+s) respectively ,which were much higher than the expressive quantity of MMP-7 gene in other differentiated adenocarcinoma tissues and the gastric carcinomas tissue without lymph node metastasis .Conclusion 1 MMP-7 gene has high expression rate in gastric carcinoma and the expression is highly specific; 2There is no significant difference in the expression of TIMP-1 gene between the gastric carcinoma and the adjacent tissue of gastric carcinoma. which indicates that TIMP has high impaction on the growth,invasion and metastasis of tumors. TIMPs, as the tissue inhibitors of MMP, can suppress the invasion and metastasis of tumors; On the other hand, as a stimulus factor for forming blood vessel epidermal growth factor (VEGF), can also promote the growth and metastasis of tumors. 3 the expressive quantity of MMP-7 gene is highly associated with the pathological differentiation and lymph node metastasis of the gastric carcinomas, which indicates that MMP-7 plays a very important role in the nephrogenesis and metastasis of gastric carcinoma.
