Objectives To investigate the expression of PTEN gene in endometrialcarcinoma , and to define the role of PTEN gene in the generation anddevelopment of endometrial carcinoma.Methods In 50 endometrial carcinmas and 15 normal human endometrium,the expression of PTEN gene was detected by reverse transcription-PCRand immunohistochemisty.Results Of the 50 endometrial carcinoma cases, the loss of expression ofPTEN mRNA was exonl-8 48.00%, exon5-9 38.00% and exon5-8 32.00%,respectively, expression of PTEN mRNA was confermed in all of the normalendometrial tissues, the relative expression of PTEN mRNA was exonl-80.52±0.35, exon5-8 0.58±0.35, respectively, it was 1.48±0.42 and 1.51±0.48 in the normal endometrial tissues. The loss and reduce of expression ofPTEN mRNA was irrelevant to histological types, FIGO stages andhistological grade in endometrial carcinomas, the loss of expressions ofPTEN protein was 60.00%, respectively, expression of PTEN protein wasconfermed in all of the normal endometrial tissues. The loss of expressionof PTEN protein was significantly relevant to histological types, FIGOstages and histological grade in endometrial carcinomas.Conclution There was a higher incidence of negative or reduce ofexpression of PTEN mRNA and protein , which indicates that PTEN geneinactivation may play an important role in the genesis and development ofendometrial carcinoma .
