Objective: To determine the expression of Activator Protein-l(c-jun /c-fos) and it's related Modulators uPA/uPAR, Integrin a , and factor VI in lung cancer specimens, try to find out the possible roles of AP-1 in the infiltration and metastasis of lung cancer , the correlation among AP-l,uPA,uPAR, Integrin a j and the correlation with tumor angiogenesis mechanism.Methods: The expression of AP-l,uPA,uPAR, Integrin a ,, Factor VI were detected in 101 cases of paraffin-embedded sections of lung cancer tissues by immunohistochemical S-P method, and 7 normal lung tissues as control. Positive intensity and microvesssl density (MVD) was counted at X 200 magnification and was examined in AOD and IOD by CMIAS2000 image analysis system The date were statistically analysed by SPSS 10.0 software package. Results:1. The expression levels of c-jun(72.78%), c-fos(70.30%) in the 101 cases of lung cancer tissues were significantly higher than those of the 7 cases control groups(/)<0.001), the common expression levels of c-jun/c-fos(63.37%) was also significantly higher in the lung cancer tissues than in normal tissues.2. The mean rank of c-jun,c-fos expression in NSCLC is 54.78 (57.55) ^ 56.18(58.19); in SCLC is 39.48 (32.38), 35.89(30.73). All these indicates that the expression level of c-jun and c-fos is significantly higher in NSCLC than that in SCLC.3. Quantitative analysis output indicates: In different types of lung cancer, the expression of c-jun and c-fos were different. Adenocarcinoma and squamouscell carcinoma> small cell lung cancer and big cell lung cancer.Adenocarcinoma has the highest positive intensity, small cell lung cancer has the lowest positive intensity. The expression levels of c-jun and c-fos in peripheral type of lung cancer was significently higher than that in central type of lung cancer4. The expression of c-jun,c-fos were remarkably correlated with differentiate degree of lung cancer(/>< 0.005). The Positive intensity was higher in tumor with high degree than those with low degree. The expression of c-jun,c-fos has no correlation with the stage of lung cancer . But when the clinical stages were combined into two groups I +11 and III+IV, the expression levels in the last group was significantly higher than in the former group (P<0.05) .There is significantly correlation between The expression of c-jun/c-fos and node metastasis of lung cancer (P<0. 005).5. The expression levels of uPA(39.60%),uPAR(37.62%) in the lung cancer tissues were higher than those of the control groups, but had no significant difference(P> 0.005).6. In different types of lung cancer, the expression of uPA and uPAR has no significant difference(P> 0.005). The positive intensity of uPAR was significant correlated with different degree of lung cancer(P<0.005). There has no correlation between the expression of uPA,uPAR and the different stage of lung cancer(P>0.005).But after the combination of clinical stages the expression of uPA,uPAR was significantly higher in stage III+IV than in stage I + II (P< 0.005). uPA and uPAR were all significantly correlated with lymphatic metastasis(P<0.005)7. The expression levels of Integrin a { (44.55%) in the lung cancer tissues were higher than those of the control groups, but had no significant difference(P> 0.005). There has no correlation between the expression of Integrin a , and the different degrees, different stages of lung cancer(P>0.005).But after the combination of clinical stages the expression of Integrin a , was significantly higher in stage III+FV than in stage I + II (P<0.005). The positive intensity of Integrin a , was correlated with node metastasis of lung cancer(P=0.000).8. MVD was significantly higher in lung cancer tissues than in the control groups(P<0.001).There were intently correlation between MVD and different gread of lung cancer(/3<0.005).9.
