Effects Of Different Routes Of Adiminstration On Acute Hemorrhagic Necrotizing Pancreatitis Of Rats

Objective: To observe the treatment effects on acute hemorrhagic necrotizing pancreatitis(AHNP) of rats through the different routes of administration from stern vein or from celiac trunk in different time.Methods: 10 rats were set as group I (sham operation),another 60 healthy SD rats were randomly divided into 5 groups(12 rats for each group),among them,the rats of group II and IV suffered AHNP with treatment administration from celiac trunk,Group III and V suffered AHNP with treatment administration from stern vein,group VI suffered AHNP solely At 3 hours after the induction of AHNP,the rats in group II and III received intravenous or intra-artial drugs, the rats in group IV and V received intravenous or intra-artial drugs at 12 hours after the induction of AHNP.The animals were sacrificed at 24 hours following AHNP.The portal blood samples were collected for measurement of serum phospholipase A2 and endotoxin,The livers and lungs were partly taken for reverse transcription-polymerase chain reaction(RT-PCR) analysis of mRNA expression of tumor nectosis factor- alpha,The pancreases were partly taken for pathologic observation,and morbidity of each group was acumulated.Results: Compared with group I ,each survey was higher in other groups; The portal serum phospholipase A2 and endotoxin and the TNF- α mRNA express in liver in group II and IV were lower than those of group III and V ;but TNF- a mRNA expression in lung in group II, III and V was lower than that in group VI and IV; pancreatic necrosis area in group II and IV was smaller than that in group III and V and VI ,the morbidity of the groups which accepted medicine from celiac trunk was lower than that of the group which accepted medicine from stern vein.Conclusions:The treatment effects on different organ injury depend on the route of adiministration in severe acute pancreatitis. Drugs delivered through stern vein induced a greater reduction of lung TNF- a mRNA expression; drugs delivered through celiac trunk induced a greater reduction of liver TNF- a mRNA expression and were more effective in reviving pancreatic tissue, it may owe to ameliorate the liver'sfunction,which may accelerate the metabolism of cytokines and keep the hemostasis of bodies.
