Objective To investigate the effects of mifepristone and gesterinone on expression of matrix metalloproteinase-2(MMP-2) cultured human eutopic and ectopic endometria. Methods Endometrial (n=10)and endometriotic(n=15) tissues obtained from patients with endometriosis during the proliferative phase were cultured with and without mifepristone(1.86xl0-6 and 1.86xl0-5mol/L), gesterinone(9.73xl0-5 and 9.73xl0-4mol/L) for 7-10days, The expression of MMP-2 were determined by semi-quantitative immuno-cyto-chemistry method. Normal edometrial samples(n=22) with matched age, menstrual cycle, nationnality served as controls. Results Mifepristone significantly inhabited the expression of matrix metalloproteinase-2 in eutopic and ectopic endometrial cell. Positive frequency of MMp-2 was 26.7% and 37.5% in the 1.86x10-5 group(P<0.05). Gesterinone also inhabited MMP-2 expression of eutopic and ectopic endometrial cells significantly. Positive frequency of MMp-2 was 20% and 30% in the 9.73 X10-4 group(P<0.05). Mifepristone did not significantly inhabited the expression of matrix metalloproteinase-2 in eutopic and ectopic endometrial cell. Positive frequency of MMp-2 was 26.7% and 37.5% in the 1.86X10-6 group(P > 0.05). Gesterinone also did not significantly inhabited MMP-2 expression of eutopic and ectopic endometrial cells. Positive frequency of MMp-2 was 66.7% and 80%in the 9.73 X10-5 group(P >0.05). Positive frequency of MMp-2 of nomal endometrial cells were not significantly less than those of ectomic and eutomic endometrium. Conclusions The inhibitory effects on the expression of MMP-2 by ectopic and eutopic edometrium may provide one of the cellular therapeutic mechanisms of mifepristone and gesterinone on endometriosis.
