As a chemical material, phosgene belongs to blackdamp, it is widely used in processing and synthesis of dye, plastic, rubber and pesticide. Besides that, it was used as a military toxic agent in the war. Thus, the United Nations Disarmament Committee defined phosgene as "double use toxic agent". At present, with the development and changes of world politics and economy, international station is not stable and terror activities have taken placed from now and then. Therefore, it is much more important to prevent, diagnose and treat phosgene injury.Some methods were used in prevention and diagnosis of phosgene- poisoning, but nothing was effective and specific on treatment of phosgene poisoning while it took place, especially when people exposed to great dose phosgene. Based on the fact that the body was brought about plenty of lipid peroxidation after phosgene injury, in this research we chose some antioxidants to determine and analyzed indexes of antioxidation in studding effects on the prevention and treatment after pgosgene-poisning, and test their dose-response relationship, and discussed the possibility of treatment by scavenging free radical.We find that the indexes of oxidation or antioxidation, such as MDA, VA, VE, GSH, GSSG and so on, changed correspondingly after the mice exposed to phosgene, which indicated the antioxidative capacity of mice decreased and oxidative injury occurred. After treating with different antioxidant, the changes ofindexes were not identical in different organs. The phenomena may be due to the targets organs of phosgene injury and the detail mechanism of the phenomena is waited for being studied in later experiments.Phosgene poisoning causes lung injury actually and there are AT- II cells injury in acute lung injury caused by many different factors. According to these facts, we explored the molecular mechanism of acute AT- II cells injury in caused by phosgene with gene chip.We can draw a conclusion from the experiment that obvious changes take place at the molecular level, such as down-regulation expressing of surfactant protein C (SP-C) and aquaporin 8 (Aqp8), up-regulation expressing of Cathepsin B (CB) and cerulopasmin (CP). According to the theory of acid-base disequilibrium of phosgene-poisoning, most genes expressing abnormally were depressed after treated with sodium acid carbonate, which implies that the treatment is effective to some extend.The primary molecular mechanism of phosgene injury was studied in this research, which provides some significative experimental foundation for researching further, preventing and treating phosgene-poisoning in effect.
