Matrine is a kind of alkaloid which plays an important role in anti-tumor, anti-infection and anti-arrhythmia. About 40%-50% of intracranial tumors are glioma, but its therapy and prognosis are not satisfying. The aim of our study is to detect the inhibition of proliferation of BT-325 and C6 by matrine, which has a potential use in clinical administration for glioma.In this study, we investigated the effects of matrine on the proliferation of human glioma cell line BT-325. MTT was employed to measure the levels of the proliferation of BT-325 cultured with matrine of different concentrations. The effect of matrine on cell cycle of BT-325 was observed by FCM. The morphological change of BT-325 cell was detected by electronic microscopy. The results showed that the proliferation of BT-325 was obviously inhibited by matrine in a dose-dependent manner. The inhibitory rate came to the peak at 32.1±1.1%, when cultured with matrine at 0.5 mg/ml. The outcome of FCM showed that 23.9% of cells were apoptosis. The morphological changes of apoptosis were also found by electronic microscopy. In conclusion, matrine can inhibit the proliferation of BT-325 and abrogate the apoptosis of the cells.Then we studied the effects of matrine on the proliferation of murine glioma C6 cell line in vitro. MTT was employed to measure the levels of the proliferation of C6 cultured with matrine. The effect of matrine on cell cycle of C6 was observed by FCM. The expression of GFAP of C6 cells was detected by immunohistochemistry in order to observe the differentiation of C6 cells. The results showed that the proliferation of C6 was obviously inhibited by matrine. The inhibitory rate came to 24.4±0.9%, when cultured with matrine at 0.5mg/ml. The outcome of FCM showed that the proliferation of C6 was obviously inhibited. The expression of GFAP was obviously enhanced in C6 cells cultured with matrine. In conclusion, matrine can inhibit the proliferation of C6 and induce differentiation of the cells in vitro.The rudiment investigation showed that matrine can inhibit the proliferation of BT-325 and C6 through inducing apoptosis and differentiation of the cells respectively, which can be a new medicine used in anti-glioma adminstration.
