Bchgrounds Diabetes is defined as hyperglycemia and non-enzymatic glycated protein, companied with disturbance of lipid and protein metabolism. Recent studies suggest that excess oxidative stress consist in diabetes patients ,and antioxidant vitamin was widly applicated in diabetes therapy .But in all of the therapy, only a -tocopherol was used. Few studys about other tocopherols isomers have been practiced. In this study, methods was developed to rapidly separate α , β, γ, δ - tocopherol isomers in human serum and to determine serum tocopherols isomers in 117 type-2 diabetes patients and 47 normal persons, and then , the regulation and ralationship beteen tocopherols isomers and diabetes were analysised.Methods The HPLC system consisted of a Inertsil silica column (100-A,3um ,4.6 X 250mm) and 7%(v/v) methyl-tert- butyl ether in n-hexane as mobile phase . Prior to HPLC, the serum sample was deproteined by ethnaol (BHTO.0625%) and the tocopherol isomers were efficiently extracted in their original isomeric conformation using w-hexane-ethyl acetate (5:1) in the presence of 2,6-bi- butyl p-methylphenol (BHT). And determine serum tocopherols isomers in 117 type 2 diabetes patients and 47 normal person, then analysis the regulation and ralationship beteen tocopherols isomers and diabetes. Results serum Y -tocopherol in diabetes patients are significant lower than controls , fasting blood glucose is significant correlated to serum Y -tocopherol levels . Cone I us i on This technique is suitable for the assay of tocopherol isomers in human serum of all range of age.There are relatively deficiency of serum Y -tocopherol in diabetes , all tocopherols isomers should be supplied in suitable proportion.
