Experimental Studies On Spermatogenic Cells Apoptosis In Rats With Varicocele And Soluble Granule Of Sheng Jing Influencing The Apoptosis

Objective: To investigate the apoptosis and the protective effect of Soluble Granules of Sheng Jing on spermatogenic function in testes of rats with varicocele by observing the apoptosis index in spermatogenic cells,the weight of testes and the changes in histology and morphology in the testes. Methods: Experimental varicocele was induced by partial ligation of the left renal vein in Wistar rats,which were divided into 3 groups randomly: varicocele group (VG),sham operation group (SOG), treatment group (varicocele + traditional Chinese medicine VMG). Apoptosis cells were detected by in situ terminal deoxynucleotityl transferase-mediated dTUP nick end labeling (TUNELE) technique. Results: (1) The weight of the left testicular in VG was far lower than that in SOG (P<0.001). There was indistinct difference between VMG and SOG (P>0.05). (2) There was certain proportion of apoptosis cells in spermatogenic cells in SOG. The incidence of apoptosis increased significantly in VG. The apoptosis index in VG was remarkably higher than that in SOG (p<0.001). There was significant difference between VMG and VG in the apoptosis index. Conclusions: The apoptosis of spermatogenic cells by varicocele may play a important role in leading to subfertility. The Soluble Granules of Sheng Jing have negtive effect on the apoptosis of spermatogenic cell. The medicine not onlsy can protected spermic production but can improve the ability of procreation of testes. The study offered a theoretic support for clinical treatment of varicocele.
