Objective: Condyloma accuminatum(CA) is a kind of sexually transmitted disease(STD), and with the progressive incidence, it is now in the second place of STD. It is human papillomavirus (HPV), a kind of virus, that leads to CA. This kind of virus live autoeciously in the nucleolus of basal cells, while a few of them in the outside of the cells. HPV inside is the main source of relapse and can exist latently in a long period. 0.5% podophyllotoxin tincture is an effective drug advocated by WHO. However it can only be used to the warts that can be seen with eyes, and if it is used extensively, serious side-effects will take place. In order to increase the concentration of podophyllotoxin in skin layer, we prepared podophyllotoxin liposome suspension. As literature described, that better effects can be achieved when some drug is incapsuled in DPPC liposome for skin use. In present ,there is not yet any reports of podophyllotoxin DPPC liposome preparation worldwide , thus we prepared podophyllotoxin DPPC liposome and evaluated its in vitro and in vivo behavior when applied on the skin.Methods: DPPC and bean lecithin podophyllotoxin liposome were prepared by rotary evaporation decreased pressure and ultrasonic. The embedding ratio was examined with fluorospectrophotometer , and the configuration of the liposome was observed by optic and electronic microscopes. Alao we carried out the stability experiments, based on those analysis methods. Next 15 porkets were chosed and separated into 3 groups,then were applied podophyllotoxin tinctrue and 2 liposome on the skin. The porkets' blood were sampled and drug concentrations were examined at different time. Chosing 5 porkets, and the places to apply drug were chosed randomly. We applied 2 liposome, podophyllotoxin tincture, bare liposome, and bare tincture to the pockets' skin. At last we made pathologic slices of the skin, and examined the podophyllotoxin storage in skin at different time according to its fluorescence by confocal laser scanning microscope(CLSM).Results: The shape of the two kinds of liposome were the same as is ever reported. Its embedding ratio is high, and they are stable. In our experiments, the podophyllotoxin concentrations in animals' blood are different, the concentrations of liposome are lower than that of tincture, and concentration of DPPC liposome descended fastest. Analysing podophyllotoxin in skin by CLSM, we found that tincture shows the pinnacle most rapidly, but it only lasted a short time of staying in skin, and the concentration descended very fast. The two kinds of podophyllotoxin liposome released drug more slowly, there was a high drug concentration in skin during a long time. The drug concentration of DPPC liposome is highest, it had a drug pinnacle descending slowly, and kept a high concentration level during a long time, the drug resorted in skin was most.Conlusion: The two kinds of podophyllotoxin liposome prepared by Ultrasonic had high embedding ratio and were stable, so the preparing method is adaptable. Podophyllotoxin DPPC liposome diffuses the drug into skin slowly, and the drug is kept in skin very long with a high concentration. Our results suggests that podophyllotoxin DPPC liposome may be much better than podophyllotoxin tincture for treating and preventing the relapse of CA.
