ObjectiveWith the development of society and the increasing rhythm of life, the morbidity associated with colon motility disturbance is rising. Now, the diseases about colon motility disturbance have drawn attention gradually. Physiological function of colon is not digestion but motility. So, the study emphasis is placed on its motility and control. Colon has a great deal of nerve plexus. The nerve plexus consists of extrinsic one and intrinsic one. Sympathetic nerve plexus and Parasympathetic nerve plexus constitute extrinsic nerve plexus. Intrinsic nerve plexus consist of sub mucous plexus, myenteric plexus and subserous plexus. Enteric nervous system (ENS) is mainly formed by intrinsic nerve plexus. ENS is relatively independent. ENS mainly modulates Colon motility. Now it has been proved that about 20 gastrointestinal peptides, which consist of gastrin, VIP, CCK, SP,SS and so on can be synthesized in ENS. According to their effect on colon motility, gastrointestinal peptides can be classified two kinds, stimulating peptide and inhibitory ones. Stimulating peptides mainly consist of SP, SS. VIP, PACAP and other peptides constitute inhibitory one. The peptides listed above represent the major pathway. So, to study the effect of some Chinese herbs on gastrointestinal peptides means much to elementary and applied study. Having processed development in the last thousands6of years, TCM has made great progress in the theory and treatment of many diseases. Though classical TCM theory doesn't discuss the motility of colon and its control, it has expounded the function of PI and WEI. It also has abundant documents and clinical experiences of treating colon motility disturbance diseases. The unique function of Chinese herbs has been realized in the world. Total Glucosides of Paeony (TGP) has been used for thousands of years. Its function is YangYinRouGan. According to indexing, many literatures have proved that TGP can modulate colon motility significantly. Large dosage of TGP can be applied to treated constipation founded by LIU Hong-Ming in clinical experience. It was also reported that TGP could advance motility of both ileum and colon directly. This effect of TGP may be due to stimulate acetylcholine (Ach) receptors and block β2 -Adrenergic receptors. Some reports suggested that ileo-colon motility of rabbit could be promoted by TGP and glycyrrhizia. To explore whether TGP modulate colon motility through improving the secreting and synthesizing of gastrointestinal peptides means not only to understand its mechanism but also to develop new medicine on modulating colon motility.Under the direction of TCM theory, this study referring to domestic and overseas research results made use of advanced laboratory technique to demonstrate TGP's actual function on traverse and longitudinal colon muscle in vitro and effect of the secreting and synthesizing gastrointestinal peptides in ENS. It also offers some theory foundation and experimental trial for developing new drug of modulating colon motility. 2. Material and methods 2.1 Material2.1.1 Animals 28 guinea pigs, weighted 400-500 gram, were offered by the Experimental animal center of Jiangsu province. Half is male.2.1.2 Medicines Total Glucosides of Paeony (TGP) was presented by Shenzhen Nanfang pharmaceutical co. Ltd. TGP was dispensed into 25% solutions. 1.0×10-3mol/l. Acecolex was gotten from the third reagent factory of shanghai. 0.5mg/ml atropine sulfate (Lot.001941) was purchased from Wuhan pharmaceutical factory. 1.0×10-4mol/1. 5-HT was bought from Shanghais branch of the Medicine company of China. 100mg/ml erythromycin (Lot.20010202) was produced Dalian Meiluo pharmaceutical factory.2.1.3 Reagent The kits of SP and VIP were purchased from Wuhan Boster biological project co.Ltd.2...
