Objective : To investigate the effects of diazepam and ketamine on ischemic neuroal damange in rats.Methods: Global ischemia was induced by 15 minutes using the method described by Pulsinelli with modifications. All rats were randomly divided into sham operation group, ischemia-reperfusion group, diazepam group after ischemia, ketamine group after ischemia, diazepam group before ischemia and ketamine group before ischemia. The brain was removed at one . three and seven days after ischemia. Histopathology test was investiged in hippocampal CA1 region. Fragment DNA was identified by terminal dUTP nick-end labeling (TUNEL) reaction.Result: (1) In ischemia-reperfusion group, apoptotic cells occur in large on day 3.after ischemia. The normal pyramical cells in hippocampus CA1 region were reduced on days 7 after ischemia . (2) The apoptotic cells in diazepam group and ketamine group were reduced. Survival neurons increased significantly. More neurons survived in diazepam group than that in ketamine group. (3) There were more neurons in pretreated groups than that in posttreated groups.Conclusion: Transient forebrain ischemia produced poptosis and necrosis .N-methyl-D-aspartate (NMDA) receptor and ?-aminobutyric acid (GABA) receptor played a role in the injury. Diazepam and ketamine partially povided neuroprotective effects. Diazepam(10mg/kg.ip) is more efficacious than ketamine( 120mg/kg.ip).
