Objective : .The aim was to study the effects of the gene expression of CD44v6 .BCL-2 and VEGF on oncogenesis and progression .and some valuable criteria for diagnosis, prognosis and biotherapy in endometrial carcinoma.Methods: The expression levels of CD44v6. Bcl-2 and VEGF were studied by S-P immunohistochemistry method in 55 cases of endometrial adenocarcinoma and 10 cases of normal proliferatic . simple hyperplastic and atypical endometrium respectively .Their relationship relating with clinicopathological features and survival was also investigated.Results: ( 1 ) The expression of CD44v6 increased progressively from normal proliferatic .simple hyperplastic and atypical hyperplastic endometrium to adenocarcinoma .whose ratios were 10%. 40%. 60% and 78.18% respectively.showing highly significant difference(P.0.001).The CD44v6 expression in 55 cases of carcinoma was inverse associated with surgical stages and metastasis of lymph nodes (P<0.05.P<0.01).\vhile no significant difference was detected in different myometrial invasion and histologic grade. ( 2 ) The Bcl-2 expression was found to be significantly higher in FIGO Gl than G2 carcinoma(p<0.05). while no significant difference was detected in tumors of different myometrial invasion, surgical stage and metastasis of lymph nodes. The expression of Bcl-2 decreased progressively from normal proliferatic .simple hyperplastic and atypical hyperplastic endometrium to adenocarcinoma .showing no significant difference (P>0.05). (3) The expression of VEGF increased progressively from normal proliferatic . simple hyperplastic and atypical hyperplastic endometrium to adenocarcinoma. with significant difference (PO.OOl).The staining intensity of VEGF expression in 55 cases of carcinoma was significantly associated with surgical stages, myometrial invasion and metastasis of lymph nodes (P<0.001.).while no significant difference was detected in different histologic grade. (4) There were statistically significant correlation between Bcl-2 and2AbstractCD44v6 , Bcl-2 and VEGF. (5) the analysis of single factor revealed that the expression of CD44v6 and Bcl-2 was significantly related with the prognosis of patients .Analyzed by Cox proportion hazard model, it showed that the prognosis was independently affected by age .surgical stage and CD44V6,and that the prognosis of patients with negative expression of CD44v6 had a shorter average survival time than positive expression group .Conclusion: the experiment suggests that CD44v6. Bcl-2 and VEGF play roles in oncogenesis and progression of endometrial adenocarcinoma. but respectively to different extent. Detection of these gene proteins has positive effects on early diagnose, malignancy determination . prognosis and treatment of endometrial carcinoma.
