Alcoholic fatty liver (AFL), is frequent in alcoholic liver disease, which is caused by alcohol abusing. The fatalness of liver steatosis that occur liver-failure or liver cirrhosis is higher than that of simple bullous liver steatosis. So it is important to study on the pathogenesis and efficiency medicine of AFL. Traditional Chinese Medicine (TCM) is getting attention by its mild self-effect and best curative effect It has been one of the directions to prevent and cure AFL.Many doctors consider that the pathological factors of AFL are Phlegm, Dampness and Stagnant blood And clearing the liver and regulating Qi, eliminating the stagnant and activating the blood, eliminating damp and dissolving phlegm are the mainly therapy. Professor LI Diangui considers that the pathogenesis of AFL is that the poison of alcohol injures stomach, then stomach fails to propel downwards, turbid pathogens is blocking, turbid pathogens is fighting with the poison of alcohol, deadlocked in middle warmer, staying at the hypochondrium.New Qing Gan Decoction (NQGD) is studied out on the pathogenesis that damp-heat and turbid toxins blocking in middlewarmer. It is mainly composed of Radix Puerariae, Hypericwn Japonicum Thunb, Folium Nelwnbinis and Rhodiola Sacra Fu Its functions are clearing heat and excreting damp, detoxifying and dissolving turbid, treating the liver and the stomach simultaneously. It has gained best curative effect in clinic. Our study plan to investigate curative effect and operation mechanism of NQGD, and to provide efficiency medicine of TCM to treat AFL, and to provide clinical and experimental basis for the theory of turbid toxins inducing disease.Part One Effect of NQGD on Preventing and Curing Rats Alcoholic Fatty LiverObjective: To investigate the curative effect and the operation mechanism of NQGD prevention and curing AFL. To approve the feasibility and practicability of Detoxifying and Dissolving turbid therapy to treat AFL.Methods: Rats AFL model was used 40 female SD rats were divided into 5 groups randomly: (1) normal group; (2) model group ( distilled spirit 3gkg-1 body weight, com oil 1ml per rat i.p. every morning; 0.9% NaCl i.p. every night); (3) Xiao Cai Hu Decoction(XCHD) group; (4) NQGD high dose group; (5) NQGD low dose group. Rats in the last three groups were i.p. distilled spirit and com oil every morning (same dose as model group), XCHD, low dose NQGD and high dose NQGD were i.p. separately every night (1ml 100g-1 weight). The experiment lasted 6 weeks. Observe pathological changes under light microscope. Test superoxidedismutases (SOD), malondialdehyde (MDA), glutathione(GSH), nitric oxide (NO)and inducible nitric oxide syntnase (iNOS)of liver homogenates and triglyceride (TG), total cholesterol (TC), total protein (TP), very low density lipoprotein-cholesterol (VLDL-C), aspartate aminotransferase (AST), glutamic pyruvic transaminase (ALT)and alkaline phosphatase (ALP)of serum.Results: (1) Morphological changes under light- microscope. Model group: liver cell contains fatty little- droplet, those pathological changes of all are moderate or severe AFL(steatosis area>50% or >75%); NQGD low dose group: fatty little lroplet scatter, NQGD high dose group: there is no fatty little-droplet in liver cell. (2) Serum TG, TC of model group are higher than those of normal group (p<0.01). NQGD groups are lower than those of model group(p<0.01). (3) Serum VLDL-C(0.54+0.14mmol/L) of model group is higher than that of normal group(0.34+0.05 mmol/L) (p<0.01). NQGD groups are lower than that of model group(p<0.01). (4) There is on difference of TP among these 5 groups. (P>0.05). (5)Serum AST, ALT, ALP of model group are higher than those of normal group (p<0.05). NQGD groups are lower than those of model group(p<0.01). (6) Liver homogenates MDA(1.79 + 0.43 nmol/mgprot) of model group is higher than that of normal group (1.04 + 0.37 nmol/mgprot) (p<0.01).
