Vascular autograft is an operational method which was usually used in surgery,but -the Vascular autografts restenosis often occurred and result in the sicks unideal therapeutic effect and many severe complication occurred, therefore.it is very important to find an ideal therapy which can inhibit intimal hyperplasia (IH) of venous autograft. This research adopted transfection p21 gene and c-fos antisense oligoneucleotid (asODN), to observe the effects of c-fos antisense oligoneucleotide and p21 genetic transfection on the intimal hyperplasia of venous autografts; to find an ideal genetic therapy of restenosis on venous autografts.20 New Zealand rabbits were selected , which were divided evenly into experimental and control group at random. The right external jugular veins were antografted into common carotid arteries in the same side. The transplanted veins of experimental group were immersed in the adenovirus-mediated p21 gene solution for 15 minutes just before anastomosis and coated with c-fos antisense olioneucleotide glue gel just after anastomosis, while the 15 minutes but nothing to be coated. The transplanted vascular sample were taken at 2 weeks after operation The intimalthickness (IT), degreeof restenosis (DR) , expression of Proliferating cell nuclear antigen (PCNA), quantity of vascular smooth muscle cells (VSMC) were determined by pathology immunohischemistry, The results had been abtained. The IT of experimental groupis 20. 59?. 43jtm , The IT of control group is 49. 53+2. 70fj.m. the DR of experimental group is 11. 50+1. 85 ,the DR of control group is 23. 34 + 5. 79 (P<0. 01). Expression of PCN A of experimental group is 32. 90 + 3. 64 and the control group is 55. 85 + 3. 92(P<0. 01). The proliferating index of VSMC of experimental group is 12. 74 + 1. 78 and the control group is 26. 25 + 5. 75(P<0. 01). The quantity of DNA of experimental group is 5. 79?. 59 and the control group is 11. 62 + 2. 19. The quantity of secretory VSMC of experimental group decreased contrary to control group by electronic microscope observation. In the conclusion, transfection of c-fos antisence oligoneuleotide and p21gene can inhibit the intimal hyperlisia of venous autografts, it is a prospective and ideal genetic therapy to intimal hyperplasia.
