The Study of the Relativity between Psoriasis and HLA-Cw Alleles in the Population of the Han Nationality in North China Objective: hi order to establish the foundation of researches on diagnosis, treatment and prognosis of psoriasis in gene level, the frequency of HLA-Cw alleles in the population of the Han nationality in North China was realized, the susceptible gene of psoriasis in HLA-Cw was sought, and the relation among the gene and the onset age, sexuality, and family history of psoriasis was further analyzed. Methods: The subjects consisted of two groups such as normal control group and psoriasis group that disease group was divided into group I (<40) and group II(>40) according to age of morbidity. All samples were analyzed for genotype of psoriasis by PCR-SSP. The direct count method was used for calculating the frequency of allele and genotype and Hardy- Weinberg test was made. The HLA-Cw alleles from control group were compared with those from psoriasis group by X1 test. )? test was used for comparing gene distribution and others between two groups.Results: DNA samples from 70 healthy people and 43 patients with psoriasis were detected by PCR with 45 specific primers and two primers as internal quality control. 20 HLA-Cw alleles and 47 genotypes hi control group versus 16 HLA-Cw alleles and 21 genotypes in patient group were found out, and the incidence of HLA-Cw alleles between 2 groups had significant difference (p<0.005). The frequency ofNo.01KY25HLA-CW*0602 in patient group and control group was respectively 44.19% and 9.28%, PO.005, RR=33.32, EF=0.86, and there was a positive correlation between psoriasis and HLA-CW*0602. The incidence of HLA-CW*0702 in two groups was separately 3.48% and 14.29%, P<0.05, RR=0.1875, PF=0.23, and there was a negative correlation between psoriasis and HLA-CW*0702. We detected HLA-CW*0704, 1502 and 1505 that were not reported by relative literatures in China.Conclusion: (1) HLA-Cw alleles were typed by PCR-SSP with speedness, accuracy and credibility; (2) The distributive characteristic of HLA-Cw alleles in normal population and patients with psoriasis of the Han nationality in North China was obtained and there was a significant difference in some genes between two groups. The gene polymorphism of HLA-Cw alleles in various races and regions was confirmed by further analysis; (3) The results showed that HLA-CW*0602 might be a susceptible gene of psoriasis and HLA-CW* 0702 could be a protective gene from psoriasis.
