Study on the effects of the end organs on the nerve regeneration after end-to-side neurorrhaphyThis experimental study was designed to investigate the effects of the end organs on the nerve regeneration after end-to-side neurorrhaphy. A sciatic model was established. The experiment included 3 parts: 1 To explore the effects of the end organs on the regenerating axons after end-to-side neurorrhaphy by means of methylamine blue stain, immunohistochemistry and electronic microscope. 2 To explore the influence of the end organs on the recovery of 1,3,4,5 spinal cord motoneuron morphologhy . 3 Qualitative and quantitative study on the changes of NGF in the accepted nerve by means of immunohistochemistry and ELISA.From the comparison between the group A, in which the accepted nerve was connected with the end organs, and the group B, in which the accepted nerve was not connected with the end organs, we found the following results: 1 The amount of regenerating axons was larger in group A than in group B, and the myelin sheath was thicker as well. The regenerating nerve fibers of group A were parallel, and were arranged orderly, on the contrary, those of group B were arranged disorderly. The recovery time of the regenerating axons'ultrastrucrure last shorter in group A than in group B. 2 The morphologic change of the lumbar spinal cord motoneuron was reversly. The number of the normal motoneurons increased continuously, but the increase was more profound in group A.(P<0.05). The area of motoneurons was larger in group B than in group A. (P<0.05). 3 The NGF was expressed in the accepted nerve at the-3-anastomosis site, and reached the peak on the 7th day postoperatively, but the quantitaty of NGF was much higher in group A than in group B(P<0.05).Conclusion: 1 The end organs promote the donor nerve to collateral sprout, accelerated the axon regeneration and help regenerating axons mature. 2 The end organs promote the morphology of the motoneurons to recover after end-to-side neurorrhaphy. 3 The amount of NGF at the anstomosis site was much larger in group A than in group B. 4 The accepted nerve connected with end organs did play a DNS(distal nerve segment)-like effect.
