Molecular tissue engineering should be a subject that combine molecular biology and tissue engineering, to study cells, tissue-inducing factors, biomaterials and the subtle relationships between them at molecular level and, finally develop better biological substitutes which would restore, maintain, or improve tissue function.Materials: In this study, bone marrow stromal cells(BMSCs) were used as seed cells of chondrocyte tissue engineering after transfected transforming growth factor(TGF)- ft , gene by LipofectAMINE. Then the biological characters of the transfected BMSCs were investigated by many methods.Methods: Recombinant pcDNAS桾GF/?, gene was constructed; BMSCs from New Zealand rabbits were harvested and cultured in vitro and transfected by pcDNAS桾GFjfl, gene by LipofectAMINE. To confirm the approval of recombinant pcDNAS- TGFjtf, gene and the results of its transfection, we used immuhistochemical staining(SABC). TheDepartment of Or a logy 4multiplication and differentiation of BMSCs were observed by flow-cytometry(TCM), transilluminating electron microscope(TEM) and other methods. The BMSCs in controlled group was transfected with pcDNA3 only .Results: The recombinant of pcDNAS- TGF/?i gene and the result of its transfection were perfect. The multiplication of BMSCs were observed at 48h after transfection in TGF /? i group, but not in controlled group. And the differentiation of BMSCs were determined to be chondrocyte after G4i8 selection. The BMSCs in controlled group were dead about 1 week after G4,8 selection.Conclusions: 1. BMSCs can be the seed cell of gene therapy. 2. This method of TGF/?, transfected by LipofectAMINE was sucessful and can be used in experimental or clinical studies.3. The multiplication and differentiation of BMSCs were significant. 4. The differentiation of BMSCs was to be chondrocyte. 5. BMSCs should be the seed cells of molecular tissue engineering.
