Objective The aim of this study is evaluate whether partial hepatectomy (PH) enhances the growth of implanted intrahepatic tumor and whether sandostatin(SS) can alter tumor growth and its intrinsic mechanism. Methods Sixty- four Wistar rats( 1 80-220g) underwent a laparotomy;48 had a left lobes of the liver PH and 16 were controls,Walker-256 tumor was implanted in the right lobes of the liver(LH).SS,SOug/kg,was administered intraperitoneally(LP) twice a day to 32 PH rats in different phase (the time of first administered is l2hour or 72hour after PH). The remaining PH rats(n= 16) and control rats(n== 16) received a similar volume of saline(NS). SS and control group was evaluated for tumor volume ~. metastases and hepatie function. at 7 and 15 days. Flow cytometry(FCM) was used to detect whether SS can induce apoptosis in tumor cells and to evaluate the proliferating ability in remaining normal regenerative hepatic cell following PH. Results groupA(control+IH+NS) show a significant reduction in III tumor volume at 7days versus groupB.(PH+ll1+NS)(p<0.0 1). groupB show a significant increase in IH tumor volume and 100% increase in metastases at 7days and l5days versus groupC~ D(PH+IH+SS)(p抸zO.01) . groupC show a significant reduction in 111 tumor volume but increase in apoptotic rate of tumor cell(p<0.01). However,this difference diminished at 14 days.(p>0.05) the hepatic fl.inction at 7days of all group had no difference (p>O.O5) ,while at l5days , groupB抯 hepatic function deteriorated signifucantly(pO.05) Conclusions (1) 4 the growth of implanted intrahepatic tumor is enhanced after PH and inhibited by SS,this suggests that factors influencing hepatic replication also affect tumor growth and metastases. . (2) SS action may mainly be caused by a direct antiproliferative effect in early stage and it can continuously induce apoptosis in tumor cell in later stage following PH. (3) SS has no significant suppression on remaining normal hepatic cell regeneration, rats has good drug durability.
