The Study On Expression Of C-kit Receptor In Acute Leukemia And It's Clinic Significance

AbstractObjective:To explore the exPression of human c-kit recePtor protCin(c-kit R, CDl l7) in acute leukemia (AL). The fOcus of the study willbe layed on value of c-ldt expression in diagnosis for acute lymphoidleukemia (ALL) and in distincting ALL and acute non-lymphoidleukemia (ANLL). The clinic features and abnormal karyotype andwith effect of chemotheraPy for ANLL and relationship ofexpression of c-kit with other expression of the antigens on theML membrane were also studed. The change of soluble c-kit R(s-kit) in AL and relationship betWeen c-kit R and s-kit was observedand Regulation of cytokine tO expression of c-kit was studed as well.Methods:Flow cytometry (FCM) was used to test respectively 24cases of ALL and 47 cases of ANLL who were all initiallydiagnosed patients to test expressim of juxtcellular c-kit R onmononeuclear cell in bone marrow. EnZyme linked immunosobentAssary (Elisa) was used to measure respectively the s-kit in initiallydiagnosed 20 cases of ALL and 35 cases ANLL. The studycombined SCF and IL-3 together with EPO to compact respectivelythe MNC from health indivicluals and patients with ANLL and thento test exPression of c-kit of above samples within period of l2hours.Results:There is no significam difference between exPression of c-kit and s-kit in heath individuals and patients with ALL.The rate ofexpression of c-kit of patients with ANLL was significantly higherthan that of healthy individuals and that of patients with ALL(P<0.0l). The rate of exPression of c-kit of M, and M, Were mosthigh, while M. and M5 were most low in ANLL. Rate of positiveexpression of c-kit of patients with higher LDH level was higherthan that of with lower LDH level. Rae of complete remission (CR)fOr patients with ANLL in the group of positive c-kit and s-kit waslower than that of negative ones.The speciticity and sensitivity of c-kit R for diagnosis of ANLL was respectively 70.8% and 85.l%,while those of CDl3 was respectively 50% and 72.3%. Rae ofCD34/ c-kit co-exPression was 4.2%. After cytokine was added,gradual elevation of expression of c-kit of heathy individualsoccurred within l2 hours, while exPression of c-kit of ANLLshowed distinct downregulation within 12 hours.Conclusion f c-kit R and s-kit R may have diagnostic value for ANLL.There is no consistence among expression of c-kit in subtypes ofANLL, which can be considered indicator for diagnosis of ANLLand can help to classify the subtype of ANLL. Expression of c-kit isrelated to s-ldt. Expression of c-kit and s-kit is correlated with levelof LDH and with abnormal karyotype and with rate of CR forpatients with ANLL as well. c-kit R is superior to CDl3 in bothdiagnostic specificity and sensitivity for ANLL. c-kit R have morepracticaI value for diagnosis of ANLL than CDl3. CD34/ c~kit co-c-kit ~expression may help to exclude ALL. c-kit R has abnomal structure and function.
