[Abstract] Objective: To assess the effectiveness of Iodine?25 interstitial brachytherapy in the treatment of hepatocellular carcinoma and to study its mechaniSm.Methods:In vitro experiments, CBRH?919 tumor cells were cultivated in RPMI?640 culture medium together with 1?25 aseptically under 37℃, 5% CO2 -95% air ,humidity-ontrolled condition. In vivo experiments, hepatocellular carcinoma were produced by innoculation of (4-6) × 105/ml tumor cells suspended in 0. 5m1 of RPMI-l640 minimum essential medium into the livers of Wister rats. 2-3 weeks after innoculation, rats were anesthetized by introperitoneal injection of 50mg/kg of sodium pentobarbital, laparotomy were carried out, tumors had diameters of approximately 1. 0 cm, Iodine-125 sources were implanted perpendicularly into the tumors. Rats were irradiated for two weeks. I-125 irradiated rats were kept in individual cages stored in steel lined cubicles of a "rat hotel? At the end of the irradiation period, rats were anesthetized and sources were removed, the sizes of tumors were measured, whole blood counts, liver function tests, pathologic examinations and electronic microscope were performed. They were compared with pre-irradiated group and control group.Results:Experiments in vitro showed obvious effect on killing cancer cells and retraining them from growing around 1?25 seeds. Acentric fragment of chromosomes were seen. In vivo experiments, after irradiation all rats were alive, the sizes of tumors were about 0. 28 cm (P<0. 05, as compared with pre-irradiated group ); All parameters of the live function tests were normal (P>0. 05, as compared with pre-irradiated group): HGB RBC and PLT changed little, but WBC LY and CR decreased obviously (P<0.05, as compared with pre-irradiated group and control group); No apparent lesions were found in the stomach, duodenum, liver, heart and lung in all rats in gross examination; Histologically ,there were little tumor cells remained and obvious fibroses in the irradiated group, while in the control group , there were a lot of tumor cells remained, lymphocytic infiltration and slight fibroses; Electromicroscopically, in the irradiated group, condensation and fragmentation of nuclear chromatin were observed, rough-surfaced endoplasmic reticulum, smooth-surfaced endoplasmic reticulum and mitochondria appeared swollen, cellular and nuclear structure of tumor cells were disrupted, but slight degeneration of tumor cells and eytolysis damaged by lymphocyte were observed in the control group.Conclusions: 1?25 interstitial brachytherapy is a safe, feasible and effective method to treat hepatocellular carcinoma and it has no significant side effect.
