Part One Culturation of SF767 cell in Vitro and construction of NC nude mice model bearing SF767 cell gliomaObjective To construct NC mice model bearing SF767 cell glioma .Methods We cultured SF767 cell in vitro .After the concentration of cell reached 1×107/ml,we implanted 0.5ml cell to 10 NC female nude mice'endermic tissue. We observed the incidence and the development of the tumor after planting and recorded the volume of the tumor .At the end of the experiment each tumor were inspected by pathology with H-Estain and DAKO PAP Kit stain for GFAP .Results Nude mice model bearing glioma by implanting SF767(5×106)cells to the front limb was successful with a rate of 100%. The survival time of mice model was 50±1.5days . H-Estain of tumor showed malignant biology character. GFAP could be inspected in all tumors.Conclusions Planting SF767 cells to NC female nude mice'endermic tissue is a good animal model for research of glioma's long-term therapy, characterized by a high incidence of tumor and stable biologyPart Two Implanting of low dose rate125I seed for mice bearing SF767 cell glioma in vivoObjective To study the remedy effection and dose-effection relationship of low dose rate125I seed administrated to nude mice model bearing glioma induced by SF767 cell.Methods 27 mice models bearing glioma were randomized into 3 groups :control group;0.3mCi seed group;0.6mCi seed group. None,0.3mCi 125I seed,0.6mCi 125I seed were implanted into the center of tumor of each group respectively. We observed survival duration of every mice .At the time of 30 days post implantation we selected 3 mice randomly in each group and killed them to get the tumors. Each tumor was inspected by pathology with H-Estain for necrosis and TUNEL stain for apoptosis. The weight and volume of each tumor were recorded .The survival time of the other mice in each group were observed.Results Implanting of 125I seed could prolong survival time of mice model, with 50.50±1.12days for control group;56.33±0.71days for 0.3mci group;62.67±1.33days for 0.6mci group. There was significant difference between each group(P<0.05) . At 30 days post implantation ,the incidence of apoptosis at the site of 2.8 Gy,5.6Gy,9.2Gy,18.4Gy were 3.91±0.95%,4.25±1.06%,15.8±1.02%,26.18±1.11% respectively .No apparent apoptosis was observed at 5mm away from the center of the tumor of control group .Significant difference existed among the site of 5.6Gy,9.2 Gy,18.4 Gy (P<0.05). Apoptosis accelerated with the dose of interstitial brachytherapy.Significant necrosis emerged when the dose of interstitial brachytherapy accelerated. ,the necrosis area percent at the site of 2.8 Gy,5.6Gy,9.2Gy,18.4 Gy were 7.13±5.42%,8.33±6.42%,26.67±14.97%,35.83±10.83% respectively。Significant difference existed among the site of 5.6Gy,9.2Gy,18.4 Gy(P<0.05) . The weight and volume of tumor were 2.38±0.15 cm3,182.6±9.2 mg for control group;1.66±0.12 cm3,100.5±3.5 mg for 0.3mci group ;0.77±0.23 cm3,71.2±3.9mg for 0.6mci group. There were significant difference between each group(P<0.05) .The weight and volume of tumor decreased when the dose of interstitial brachytherapy accelerated.Conclusions Apoptosis and necrosis may both be main mechanism of interstitial brachytherapy with low dose rate 125I seed . Interstitial brachytherapy could prolong the survival time of mice and decrease the weight and volume of tumor.Part Three Operation and intra-operative low dose rate 125I seed permanent implanting for recurrent malignant gliomaObjective To assess the effect of intra-operative low dose rate125I seed permanent implant for recurrent malignant glioma.Methods 6 recurrent malignant glioma patients underwent reoperation with microscope and intra-operative low dose rate 125I seed permanent implanting. All patients were followed–up.Results Up to June 2006 ,the longest follow-up period was 38 months , with the shortest 10 months after reoperation . Only one patient died at 20 months after reoperation because of recurrence, with 5 patients survived. No significant difference was found about the function of liver,kidney and thyroid gland after implanting. KPS scores of 3 months after reoperation showed no significant difference, compared with those before reoperation.No significant radical necrosis was found on CT after reoperation.Conclusions Intra-operative low dose rate 125I seed permanent implanting was an active and effective remedy for recurrent malignant glioma.
