All along, the shortage of donor organs is a important factor to hamper thedevelopment of allotransplantation, and this quantitative incompatibâ…¡ity of donor andrecipient is more critical at present than before. As the most promising alternative tothe current organ shortage, xenotranspIantation(XT) was resumed in late l980's andmany important developments in this field have been achieved and several clinicalxenograft tria1s were reported. However, with extensive researches, people graduallyknow that many immunoIogicaI and pathophysioIogicaI barriers existing in speciesneed to be overcomed l and before this, any clinical application will be b1indfold.Thus,in recent years, the focus of XT is fundamental researches.Hamster-to-rat liver xenograft model is a standard small animal model to studyimmunological and pathophysiological problem of liver XT moreover, this model hasspecial value for seIecting immunosuppressive drug.T â…¡ is extracted and refined from the root of Tripterygium Wilfordâ…¡Hook-F(TWHF).In recent years, Tâ…¡ have been applicated to allotransplantation as aimmunosuppressive drug and many successful experiences have been achieved,howevef, in XT the effects of T â…¡ aren't definite and eXtensive researches arenecessaryThe purpose of this study aimed at constructing hamsterto-rat liver xenograft modeland investigating the immunosuppressive effect of T â…¡.MATERIALS AND METHODSl. Female Golden hamsters and inbred male Wistar rats were used as donors andrecipients, respectively. Orthotopic liver transplamation was perfOrmed byKamada's cuff technique with some modifications. Animals dying<3 days aftersurgery were exc1uded from the study;2. Experimental groups were divided into two groups: (I ) (n=l5),Untreated control,(â…¡ ) (n=l5), T â…¡;3. Observed physical sign changes of postoperative rats. Measured the body weightof the recipients in two groups every other day posttransplantation. Liverspecimens of 2hr, 72hr and death of the recipients postoperatively were collected,respectively and examined by pathology;4. Statistical analyses according to survival time of recipients.RESULTSl. 30 hamster to rat orthotopic liver xenografts were performed in this study. Theoperative success rate was 80%. The model proved reliable for studies of liver xT2. The survivals of untreated control and T â…¡ groups were 7.l i0.35 and 7.2 i0.52days,respectively, and there were no significant differences in survival indexbetween two groups.Tbe result suggested that as monotherapy,30mg/kg/day Tâ…¡couldn't prolong recipient survival.3. The conjunctival congestion incidence of untreated control and T lI groups werel00% and 3l%, respectively, and there were significant differences in this indexbetween two groups. The mean emergence time of conjunctival congestion ofuntreated control and T lI groups were 4.73 f 0.47 and 5.75 l 0.50 dnys,respectively, and there were significant differences in this index between tWogroups. The results suggested that conjunctival congestion couId be regarded as asign of rejection and T â…¡ could relieve it.4. At days +l, +3, +5, +7 posttransplam8tion, the body weight of untreated controlrecipients were l50.82f9.87,l49.55t l0.05,l49.09i 10.l0 and l48.00l l0.00respectively, and there were no significant differences in this index. The sameresult could be found in T â…¡ group. Those suggested that no change of the bodyweight of the recipients of this model could occour in this period of survival and30mg/kg/day T â…¡ didn't bring change of it.5. Histology ORejections were not seen in both untreated group and T â…¡ group at 2hr after sergery. @Rejection was obvious in untreated group at 72 hr after surgery,while mild in T â…¡ group. @Although rejections were severe in both groups at thedeath of the recipients, it seem slighter in T â…¡ group than in untreated group. Theresults of histology as above indicated that T â…¡ could inhibit the rejection of this...
