Multi-infact dementia(MID) is a kind of disease which cerebral tissue and intelligence are damaged by iachemic neuronal necrosis in cerebral infarct or embolism. The focus is in brain, not convinence to study. A perfect animal model is necessary for deeply inquirement. This test will improve the model and prove it from behaviors pathology and the content of amimo acid. Wulongdan has clinically been used to treat MID. We hope our research can provide theoretic and experimental evidence for its exertion. Method: The rat model team are injected the dry-blood embolus from intracarotid. Cerebral embolism抯 size~ position and volume are observed with TTC ~,. HE staining; NSE s-i oo protein are tested by inirnunohistochemical staining. The change in neuro-supermicro structure is observed under electronmicroscope. Amimo acid抯 content is measured by high efficiency liquid phrase. The level of learning and memory in rats is tested by Y-maze, and MID rats are tested with Chinese Medicine梂ulongdan(WLD). Results: White ischemic fields are found in cerebral tissue by TIC staining and the embolism position in cortical substance抯 frontal part.. temporal part and hippocampus of brain by HE. Neurons become deformed gathered and decreased. NSE. 5-100 protein are generously showed. Ischemic.. necrosis changed is occurred in neuronal supermicrostructure. The content of excited amimo acid(EAA) ~ inhibited amimo acid(IAA) aromatic amimo acid(AAA) and other neural acid decrease. The function of learning and memory dropped(P<0.05) -3- and are increased after taking WLD. Conclusion : An improved method is created by injecting dry-blood liquid into rat抯 intracarotid for MID model. All results prove that our model method is better in stability, higher in survival and success ratio than old ones. WLD can improve the level of learning and memory. The mechanism may be related with enhancing the content of EAA and IAA in brain.
