| On the basis of toltrazuril solubility in oils,emulsifiers and cosurfactant,s,the suitable oil,emulsifier and the cosurfactant were preliminary selected,with the help of pseudo-ternary phase diagram,the nanoemulsion ingredients were screened and optimized,the nanoemulsion appearance ,the shape of nanoemulsion particle and the size distribution were assessed, by centrifugation, keeping nanoemulsion sample review test and the constant temperature accelerated test,the physical stability was also assessed.on the basis of the blank nanoemulsion ,the toltrazuril nanoemulsion was prepared,the quality assessment and the physical stability were also finished.the treatment effectiveness of the toltrazuril nanoemulsion and drug combination was assessed by the Pharmacodynamic experiment ,which the chicken were infected in laboratory.it was advantage in improving of toltrazuril nanoemulsion treatment effectiveness and increasing the toltrazuril dosage forms,at the same time,it also provided the science evidence for toltrazuril nanoemulsion using in the poultry breeding.The oil were chose from some kinds,for example,the oil were marked O1,O2,O3,O4,O5. As well, the emulsifiers were selected from some that were also markedS1,S2,S3,S4,S5. By the same way, the suitable co-emulsifier was picked from C1,C2,C3,C4,C5.the toltrazuril dissolubility in oil,emulsifier and cosurfactant were tested,based on the primary data ,the nanoemulsion was tried to be made . with the help of the pseudo-ternary-phase diagram, the emulsifier and the cosurfactant were mixed as the weight ratio, then the mixture and the oil were mixed together following the weight ratio (1:9,2:8,3:7,4:6,5:5,6:4,7:3,8:2,9:1),with the mixing, the deionized water was added into them.at last, The prescription of nanoemulsion was choose.The prescription of nanoemulsion was made up of S4,O2,C5. The nanoemulsion area of the pseudo-ternary-phase diagram was the maximum as the Km value was three , when the Km value was three and the weight ratio between the mixture and the IPM was four,the quality of the nanoemulsion was best. The mean diameter of blank namoemulsion was 42.99nm, the size distribution was centralized and the fluidity was in good condition.the nanoemulsion was proved that the quality was good by centrifugation, constant temperature test and others.When the toltrazuril nanoemulsion was prepared,the toltrazuril was added into the mixture which was made up of the emulsifier and cosurfactant,both the mixture and the oil were mixed as the weight ratio (1:9,2:8,3:7,4:6,5:5,6:4,7:3,8:2,9:1),with the deionized water dripping.when the slight opalescence appeared,the toltrazuril was successfully prepared. the toltrazuril nanoemulsion appearance ,the shape of toltrazuril nanoemulsion particle and the size distribution were assessed, by centrifugation, keeping nanoemulsion sample review test and the constant temperature accelerated test,the quality of toltrazuril nanoemulsion was also assessed.on the basis of the toltrazuril nanoemulsion.test,the results showed that the appearance of the nanoemulsion was transparent,the partical of the nanoemulsion was spherical through transmission electron microscope,the mean diameter was 77.88nm,so the quality of toltrazuril nanoemulsion was stability.The pharmacodynamicses of toltrazuril nanoemulsion and the drug combination were assessed,the chicken were infected by the coccidium in laboratory,the eating,drinking and the hen dung were observed, when the chicken pulled out the blood,we use the prepared drugs to treat the chicken. At last , the treatment effectiveness was assessed by the oocyst value, survival rate, anticoccidial index and the others.The results showed that the anticoccidial index of the toltrazuril nanoemulsion was 171.61,the toltrazuril nanoemulsion treatment effectiveness for the chicken coccidiosis was significant.The quality of toltrazuril nanoemulsion is stability,it is convenient to be used and the treatment effectiveness is significant.
