| Intramuscular Fat Content affects tenderness, juiciness, flavor, morphology, and so on. Muscle fiber types and intramuscular Fat Content are associated with muscle tenderness. PPARy2, PGC-1αand MEF2C gene can affect intramuscular fat content and muscle fiber types, thereby affect beef quality.In this study, we used Simmental cattles of different developmental stages as testing materials,measured the intramuscular fat content and shear force of longissimus in different age stage of 6 months,12 months,24 months and 36 months,researched the gene expression pattern of the PPARy2, PGC-la and MEF2C using fluorescent quantitative PCR method, and analyzed the correlation of the expression levels of three genes, intramuscular fat content and shear stress. The main results are as follows:With age increasing, the longest muscle of Simmental Cattle intramuscular fat content and shear strength are gradually increased, and intramuscular fat content is negatively correlated with shear force, correlation significant (P<0.05), except the 6 months of age.PGC-la is widely distributed in abdominal fat, kidney, back fat, heart, liver, spleen, soleus, gastrocnemius, longissimus dorsi and semitendinosus muscle, and expression pattern is different in different ages and different organizations. The relative expression of PPARy2 of different fats is significantly higher (P<0.01) than that of the different types of muscles, the expression pattern showing a continuing upward trend with age and a sharp increase of 36 months old in the fats and muscles. The expression of MEF2C is low in four muscle.Correlation analysis showed that, in the 6 months,12 months,24 months and 36 months, the expression of PGC-1α, PPARy2, MEF2C and the intramuscular fat content, shear force of longissimus are not related significantly (P> 0.05). |
