In this experiment, we used transmission electron microscope (TEM), immunofluorescence combined with flow cytometry (FCM) technique to study the effect of cadmium on spermatocyte and spermatozoon apoptosis in testis, as well as the quality of mature spermatozoon in epididymis by artificial water contamination. The purpose of present study was to supplement the basic theory of reproductive toxicology on cadmium, and to lay the foundation for the later research on the expression of spermatocyte apoptotic gene and tyrosine phosphorylation protein. The results showed that the amount of spermatocyte apoptosis in experimental groups mouse was remarkably higher than the control group. The ultrastructure of spermatozoon in the period of apoptosis showed the phenomena such as mitochondrion vacuole, karyotheca invagination, chromatin margination and karyopyknosis. All above indicated that cadmium can cause spermatocyte apoptosis in mice easily. The rates of early spermatozoa apoptosis in experimental groups were higher than the control one. Meanwhile, the rates of active spermatozoon (75.1%) were lower (P<0.05). Among all, the survival rate of spermatozoon in the 0.10mmol/L CdCl2 treated sample was remarkably lower than the control group and other experimental ones, and the rate of early apoptosis (22.6%) was the highest. The DNA breakage rate of testis germ cel(l18.2%) and epididymal serm (26.5%)in high-dose group were significantly higher than the controlled group (3.3%,5.6%)(P<0.05). The DNA breakage rates of testis germ cell were lower than epididymal sperm in different groups. With the increase in the dosage of Cadmium additives, within the apoptosis of mouse testis germ sell and epididymal sperm gradually increased . From the result we can conclude that the dose of cadmium definitely has something to do with the quantity of apoptosis and damaged spermatocyte DNA in mice germ cell.
