The seeds of Cichorium intybus.cv.Puna were irraditiated by 60Co-γ-ray,chemical by NaN3 and were spaced at different dose. First, the seed germination and seedling characters were surveyed in laboratory. Second, the seedlings were measured by physiological and biochemical characteristics. Then, the seedlings were measured by physiological and biochemical characteristics. At the last, this paper reported study of the chromosome karyotype of Cichorium intybus.cv.Puna. The resluts showed the chromosome karyotype of Cichorium intybus.cv.Puna. Separately evaluate the effect of three kinds of mutation breeding, Main findings of the study are as follows:1. The results indicated that 60Co-γray,NaN3 and space mutagenesis of Cichorium intybus.cv.Puna seed germination with different degrees of stimulation effects. Which the germination index is:chemical mutagenesis> irradiation mutagenesis>Space Mutation; Main root length:Space Mutation>chemical mutagenesis>radiation mutagenesis; Root fresh weight:Space Mutation>radiation-induced> chemical mutagenesis; Stem and leaf fresh weight:Space Mutation>chemical mutagenesis> radiation mutagenesis.2. The results indicated that 60Co-γray,NaN3 and space mutagenesis make Cichorium intybus.cv.Puna seedlings showing different degrees of stimulation effects on physiological and biochemical characteristics. Which the content of chlorophyll,the accumulation of free proline and POD activity are:radiation-induced accumulation of seedlings greater than space mutagenesis and chemical mutagenesis; Which the content of soluble protein,SOD activity and the accumulation of MDA are:space is greater than the effect of chemical mutagen and radiation-induced mutation; Which the content of soluble sugar is:chemical mutagenesis have the largest accumulation, it is higher than the radiation-induced mutagenesis and space.3. The results indicated that 60Co-γray,NaN3 and space mutagenesis make Cichorium intybus.cv.Puna seedlings showing different degrees of stimulation effects on physiological and biochemical characteristics. High doses of radiation has played a significant inhibition on the growth of chicory; Chemical mutagenesis of various doses little impact on chicory; Space mutagenesis materials have the best overall performance.4. This paper reported study of the chromosome karyotype of Cichorium intybus. cv.Puna. The resluts showed that the chromosome karyotype of Cichorium intybus.cv.Puna is 2n=2x=18=6m+12sm. The scope of the relative length is 7.04%~14.53%.The average arm ratio is 1.85. The ratio of the longest chromosome and the shortest chromosome is:2.32. The arm ratio of each chromosome is less than 2. Belong to the 1B type of symmetry. Belong to a relatively primitive plant species in the evolution system.5. In the mutation of three ways, although the low dose of radiation mutagenesis and high dose of chemical mutagenesis are in varying degrees, on the Cichorium intybus.cv.Puna physical activity played a catalytic role, but the space mutation, on the Cichorium intybus.cv.Puna physical activity the greatest impact, the specific performance of the higher soluble protein content, higher SOD activity, shorter reproductive cycle; its tall plant morphological characteristics, number of branches per plant more than the base diameter large number of reproductive branches and flowers and more; these characteristics make the best effect mutation, can be used as breeding material for further breeding center.
