Study On The Technical System Of Tissue Culture And Rapid Propagation Of Cynanchum Wallichii Wight

This paper researched on the tissue culture system of the wild plants Cynanchum wallichii Wight, for exploring the application value in gardening and better germplasm resources protection.The study of tissue culture technique and the better medium for various stages were as follows:1,The best season to draw materials was April,in which the germination rate using stem,leaf and internode as explants were 92.73%,90.57% and 96.80%,and the contaminated rate only were 28.03%,27.80% and 19.37% respectively.2,Initial culture:Initial medium of stem was MS+6-BA1.0mg·L-1+NAA0.05 mg·L-1+IBA0.05 mg·L-1 in which the germination rate was 93.4%, the shoot forming index was 1.8 and the callus induction rate was 89.23%.Initial medium of leaf callus were MS+6-BA 0.5 mg·L-1+ NAA 0.01 mg·L-1,in which the callus induction rate was 100%;Initial medium of internode callus were MS+6-BA 2.0 mg·L-1+NAA 0.01 mg·L-1+2,4-D 0.05 mg·L-1,in which the callus induction rate was 99.57%.3,Multiplication culture:Proliferating of stems cultivation was:MS+KT1.0 mg·L-1+NAA 0.1 mg·L-1+IBA 0.1 mg·L-1,,in which the multiplication rate was 4.17 and the multiplication of efficiency seedlings was 3.37.Proliferating callus of leaf was obtained following cultivation on MS+6-BA 2.0 mg·L-1+NAA 0.2 mg·L-1+ 2,4-D 0.05mg·L-1.The proliferating rates was 4.83.The best culture medium for callus of leaf differentiating was MS+6-BA0.5 mg·L-1+NAA0.4 mg·L-1.The regeneration rate was 92.73%.4,Efficiency seedling culture:the best medium for efficiency seedling was MS without any plant growth regulator.5,Rooting culture:the medium with the best combination of plant growth regulator was 1/4MS+IBA 0.5 mg·L-1,with the rooting rate 99.53%, the quantity of roots 6.70, and the length 3.90cm.6,Forging seeding transplanting:The optimized transplant medium for Cynanchum wallichii Wight was vermiculite, perlite and riversand mixed by 1:1:2 with the survival rate 66.7%.
