| As one origin of the Brier grape(Vitis davidii Foex), Hunan province distribute rich resources of wild germplasm. In this research, in order to understand their genetic relationship, emploed the observation of Botanical characters and analysis of SRAP, we distinguished Genetic diversity of 21 Brier grape resources from different areas in Hunan Province. The main results are following:1. Encoded the various types of Brier grape by Galet's coding According to leaf characteristics, and take a cluster analysis. We found Gaoshan NO.2 is the same type of Muyexi Brier grape; white Brier grape quite different from other types.2. Cluster analysis results of Brier grape fruit character Showed that Significant differences exist in white grape between other types3. In this study, there are high impurity in the total DNA extracted from Brier grape leaves, these impurities are polysaccharides, polyphenols, protein substances. Use the ethanol instead of Isopropanol to precipitate DNA from Brier grape Leaves. Found that the improved DNA methods, impurity content lower. Also found that more than 5 hours or overnight precipitation, precipitation will occur yellowing, browning, but only 4 to 5 hours sedimentation, flocculent precipitate was translucent. The experiment found that by improved CTAB method DNA extracted to meet the requirements of SRAP-PCR4.. Seven pairs of primers from the 84 pairs of primers were selected, which amplified 61 clear bands and 7 article difference bands, Polymorphism rate was 55.7%. UPGMA cluster analysis and principal coordinates analysis show that: there is 7 types of Vitis davidii Foex In Hunan.
