Now the earth which people's survival and development relying on is facing the degeneration of zoology environment day by day. The environment pollution hidden trouble caused by scaled stock raising and the consequence caused by misusing feed additives lead people to pay more attention. To study and develop no or low public hazard zoology silage as soon as possible is to solve the contradiction between development of stockbreeding and environment pollution,it takes on very profound and lasting meaning for assuring continuant development of stockbreeding and forage industry. Our country is a big agriculture country, but the use ratio of crop straw is still quite low in hometown. A great deal of straw was burned in the field, it caused not only air pollution and zoology environment destroy but also natural resources waste. The succulence or straw made into silage forage can be preserved for a long time, and the inhere juice and nutrient can also be locked. The silage forage being fragrant and very tendemess is easy for the digest for domestic animals. Absorption ratio of sundry nutrition is higher than dry forage. One of the most important methods of modern silage technology is to use silage additive. There are many relative reports in foreign country, the study was all round using pasturage and whole mealie as raw material to the silage, but in our country most of raw material is mealie straw. So carrying out study of mealie straw silage additive has important practical meanings aiming at the specific circumstances of our country. This subject is mostly to dispart lactobacillus from nature environment and sieving befitting lactobacillus served as main body of biological silage additive which be fit for new type mealie of Liaofeng–2. Then do optimize the conditions of growth and fermend.The experiment mainly conclude three parts:(1)The experiment of isolating strain: Separation, isolating and purification some strains of lactobacillus through samples of cornstock ,nearby soil , sour cabbage, soil around flocks and herds breed aquatics field and dejection flocks and herds in Gaowan region of Fushun. We found out that capacity of producing lactic acid of strain of GY-3 was the best through contrast experiment, then we confirmed strain of GY-3 as experiment strain. And we identified the strain, this strain is Streptococcus.(2)The liquid fementation feasibility study of the strain of GY-3Most of Lactobacillus is facultative anaerobes or microaerophile, and acetyformic acid can be transformed into lactic acid under anaerobic condition. So, it is important to control oxygen to produce lactic acid through lactobacillus ferment. Through four ferment ways of contrast experiment, we found out that ferment way ofⅠis the best, and 14h is optimal control time point. Effects of carbon resource and nitrogen resource were studied, the optimal carbon resource and and quantity of carbon resource and the optimal nitrogen resource and quantity of nitrogen resource are respectively glucose , 2%, bean cake, 2.5%. The orthogonal test was applied to optimize other component of the culture medium: juice of tomoto, dipotassium hydrogen phosphate and disodium hydrogen phosphate, which are respectively 2%, 1‰, 2‰.On the optimal culture medium, the initial pH, cultivate temperature and quantity of inoculate are studied, the optimal fluid ferment conditions are respectively 6~7, 38℃,2%. Based on the foundation of optimal culture medium and fluid ferment conditions, we me set out the growth curve of GY-3 and then determined that the optimal harvest time is 15h.(3) the applying effect of Gy-3 in silageUnder the optimal operation conditions, GY-3 is largely cultured and made into 2×108 bacterial suspension for following study. Changing ratio between lactobacillus and distilled water, with brown suger, urea, sodium chloride and cornstalk being respectively 0.8g, 0.4g, 0.24g, 80g, the applying effect of GY-3 added into silage forge was researched. The experiment results showed that the optimal addition amount is 1.875ml/g(silage) and can improve the quality of silage.Though GY-3 strain have a good effect of controlling mold, the degradation rate is to be improved. So the further study on compound utilization of GY-3 and celluase is to develop.
