In this study, the genomic DNA of the arecanut extraction method was improved, optimization of the arecanut RAPD reaction system, established the best RAPD amplification system, analysis the arecanut germplasm resources of Hainan genetic diversity. With graphics and tables to reveal the relationship between genetic diversity of Hainan yellow leaf disease of arecanut. The main results of this study were as follows:1. This experiment optimized the RAPD reaction conditions of areca RAPD-PCR reaction system.25μL reaction system,it contained 1×Buffer, primer 1μmol/L,Mg2+2 mmol/L,dNTPs 0.2 mmol/L,Taq enzyme 1 U,genome DNA 50 ng. The process of PCR were 94℃for 4 min,94℃for 45 s,34℃for 60 s,72℃extending for 90 s,40 cycles,72℃extending for 10 minutes.2. From the 80 RAPD primers screened 10 primers. Using 10 primers to 91 samples for RAPD analysis. Total of 101 fragments amplified, fragments between 600bp-3000bp.78 polymorphic fragments, the Polymorphism of the arecanut groups is 77%.Hainan arceanut cluster analysis of results show that the genotype of Hainan arecanut is not the same, the individual is between a certain polymorphism.Non-existence of homozygous groups. Genetic relationship between arecanut's groups are close, but the genetic diversity of populations rich. Hainan arecanut cluster analysis of individual and group analysis results were similar, only slightly lower similarity coefficient.3. Arecanut on the main plant in Hainan in 20 counties and cities 74 samples. Cluster analysis showed that arecanut samples collected in the same area similarity coefficient between the concentrations of about 0.93. Hainan nut genotypes between individuals are inconsistent with certain polymorphisms. There was no homozygous group. Arecanut in the same area of the genetic relationship between individuals are close, but the genetic diversity among individuals rich; different parts of the betel nut similar to the coefficient of 0.93 the following, individual differences are significant.4. Hainan, Yunnan, India, Cambodia arecanut samples and samples of the areca triandra cluster analysis showed that the areca triandra and arecanut similarity coefficient is 0.38, indicating areca triandra and distant relationship between arecanut; similarity coefficient of 0.78 in the group was about arecanut divided into two categories, the first category includes Yunnan and HainanⅠ, India, Cambodia samples. HainanⅠand YunnanⅠwhich close relationship, India, Cambodia sample which close relationship.the second category, including YunnanⅡ, HainanⅡsample, the similarity coefficient of about 0.8. 5. Hainan arecanut phytoplasma disease morbidity area and not epidemic areas Cluster analysis diagram analysis showed that when the similarity coefficient of 0.79 arecanut can be divided into two categories.There are two types of distribution in samples of non-infected, however groups of two types of arecanut exist some differences in the onset of disease time and the onset of disease extent. The first type of onset arecanut groups, onset earlier and severe. The second type contains more non-disease arecanut groups.
