In order to explore the advanced technology of grass production, Lactuca indica L.cv.Mengzao was taken as raw material referencing the extraction quantity of the leaf protein, crude protein and the crude protein content of leaf protein by means of comparison the acid heating method, alkali heating method, salting-out method and organic solvent extraction to determine the modest method and craft condition. The best extraction about Lactuca indica L.cv.Mengzao of leaf protein, the change of crude protein and amino acid content of leaf protein and the method of dry and store have identified in this thesis. The conclusions are as follows:①The high temperature heat treatment and acid environment relatively suit the extraction of Lactuca indica L.cv.Mengzao's leaf protein. The extraction of crude protein from acid heating is clearly higher than the three other extraction methods. The proper extraction condition is 90℃, pH=5 and 4min condense time. In the aspect of the extraction of Lactuca indica L.cv.Mengzao's leaf protein, the effects in four types of extraction are as follows: Acid heating>alkali heating> organic solvent extraction> salting-out method.②The Lactuca indica L.cv.Mengzao should be cut in 50~120cm height to extract the leaf protein.③The drying methods of Lactuca indica leaf protein extraction in the order of crude protein are as follows: 65℃constant temperature>drying>vacuum drying>dry. The extraction of amino acid content of leaf protein after drying are as follows: drying>65℃constant temperature> vacuum drying> dry. The influence of drying method and 65℃constant temperature method are least for amino acid content of leaf protein. It is most economic reasonable by cooperating two methods.④By measuring the crude protein content and amino acid content, the loss of protein content are least in brown paper leaf bags at 0℃.
