Endometrium was the mucous tunic of uterus, which could cause serious harm and economic loss to animal husbandry. At present, there are many ways for the treatment of cow endometritis, such as hormone therapy, antibiotic therapy etc. The research methods and levels were mainly to observe the effect, its mechanism was still not very clear. In this study, the uterine protection function of the traditional Chinese medicine compound preparation"Yimu Shenghuasan"and the molecular mechanism of interaction between single herbs were investigated by in vitro and in vivo experiments.In vitro study, dairy endometrial cells were isolated by collagenase digestion method and treated with gradient concentration of the traditional Chinese medicine compound preparation"Yimu Shenghuasan",Radix Glycyrrhizae, Angelica, Motherwort, Honeysuckle for 24-72h respectively, and the expression of cytochrome P450 in dairy endometrial cells was determined by Western Blot. The results as follows, (2-4)×108 cells per cow were obtained and the viability was about 95%. Dairy endometrial cells grew well, had a good morphology and could be used for investigation of drug metabolism. The dairy endometrial cells induced inflammation by lipopolysaccharide (LPS). The culture supernatants were collected in different period, and the contents of TNF-α, IL-1βwere determined by ELISA. The results showed that 100 ng/mL LPS was the optimal concentration to induce the inflammation of cultured endometrial cells. CYP450 in dairy endometrial cells had no induction with the traditional Chinese medicine compound preparation"Yimu Shenghuasan"in 24h, for 48h the expression of cytochrome P450 in cow endometrial cells increased gradually by Western blot, when the drug concentration reached 5000μg/mL, the expression of CYP450 of endometrial cells had a decreasing trend; reached 10000μg/mL, the expression of CYP450 disappeared;at 72h, the expression of CYP450 became higher but later decreased. The best dose was 2000μg/mL, and the best time was 48h. When put the monomer composition Radix Glycyrrhizae, Angelica, Motherwort, Honeysuckle into the inflammatory cell separately, they had the different induction of CYP450, and the effects are not very ideal.In vivo study, in a dairy farm, healthy dairy cows and dairy cows with endometritis were selected as experimental animals, then blood samples from external jugular vein were collected at the 0, 3th, 5th, 7th day. SOD activity and serum MDA content were surveyed by clinical pharmacodynamics test, to observe the effect of the traditional Chinese medicine compound preparation"Yimu Shenghuasan"on cow endometritis. The results showed that"Yimu Shenghuasan"decreased the content of MDA. It also could enhance the antioxidation ablility and decrease the peroxidation degree of the lipid.It was concluded that the compound emulsion had an obvious effect on dairy cow endometritis.
