Background:Differential analysis of gene expression profiles in endometrial cells of dairy cows under high concentration of palmitic acidObjective:This study through the high-throughput sequencing technology comprehensive analysis of high concentration of palmitic acid(PA)under the condition of different cow endometrial cell function gene expression,so as to explore the effects of high concentration levels of palmitic acid on gene expression of bovine endometrial cells,and provide a theoretical basis for further study on effects of palmitic acid in dairy cows endometrial cell function and to explore the relationship between the cow ketone disease and other metabolic diseases and reproductive disorders.Methods:First of all,we first measured the relative survival rate of palmitic acid at different concentrations by the method of MTT in vitro culture of endometrial cells.And then,using high-throughput sequencing method,we studied the gene expression profile of endometrial cells of dairy cows affected by palmitic acid.Finally,using the quantitative PCR technique to verify the differentially expressed genes,reconfirmed the effect of palmitic acid on the gene expression of endometrial cells.Results:Genes were characterized as differentially expressed,and 188 up-regulated and 178 down-regulated genes were involved in physiological process,such as inflammatory cytokine signaling pathway,B cell receptor signaling pathway,HIF-1 signaling pathway,insulin secretion and other physiological processes were selected for validation by a quantitative real-time polymerase chain reaction(qRT-PCR)analysis,the qRT-PCR results are consistent with the general expression patterns of 9 genes from the Illumina Sequencing.The result showed,under high concentration of palmitic acid effectting on endometrial cells,these genes expression were associated with inflammation and metabolism,which was up-regulated on TGFB1,IL6,and which was down-regulated on VAV1,CCL28,BLNK,TGFB3,IGF1R,GPX1,SOD2.Conclusion:The high concentration of palmitic acid can be caused by bovine endometrial cells differentially expressed genes related to inflammation and oxidative stress pathways,suggesting that palmitic acid may be the important factors to reduce the reproductive performance of dairy cows in postpartum dairy cows induced inflammatory disease of the uterus.
