This experiment uses immunochemistry and real-time quantitative PCR to detect the localization and expression of BMPRII in goat COCs during in vitro maturation.We further approached the influnce of BMPRII on oocyte,in order to provide a theoretical basis for the biological function of BMPRII. In addition,the test optimized the system of IVM and IVF of goat oocyte, so as to supply the experimental evidence for the production system of goat embryos in vitro.The results showed that:1. BMPRII mRNA was expressed in the whole process of in vitro maturation of oocytes. It showed the tendency of increase at first and its expression peaked at 12h of IVM,then decreased, in accord with cumulus expansion time.2. BMPRII shows strongly positive signal in follicular fluid and oocytes, and in follicle cells of all levels of follicles, BMPRII shows relatively weakly positive signal.3. The findings suggest that BMPRII may play a important role in the process of goat oocyte maturation and cumulus expansion, and further proof that BMPRII is also the receptor of BMP15 and GDF9.4. Different concentrations of serum (10%,15%, 20%) and maturation time (24 or 27 hours) have little effect on the in vitro maturation of oocytes (P>0.05); ovarian diameter of oocytes has significant effect on it (P<0.05).5. Oocyte maturation time (24h,27h) has no significant effect on fertility rate(P>0.05); the diffrence is extremely whether there is cumulus oophorus or not had extremely significant effect on the in vitro maturation of oocytes (P<0.01); the diffrence is significant whether removing cumulus oophorus or not (P<0.05).
