| Cashmere is in the inner of goat hair, the product of secondary hair follicles, and has soft, smooth, value and other characteristics, is a high textile material. The fine quality of cashmere has a close relationship with its structure protein including keratin and keratin-associated protein. The content and structure of keratin was the relatively stable in different kinds of goats hair follicles, while the keratin-associated protein (KAP) was changed greatly. In order to study the relationship between HGTKAP gene expression and cashmere fineness, the improved mechanical separation was used to obtain the primary and secondary hair follicles, and quantitative real-time RT-PCR(qRT-PCR) was firstly used to detect the levels of KAP7.1 and KAP8.2 gene expression in primary and secondary hair follicles of Liaoning New-breeding cashmere goat; semi-quantitative RT-PCR was used to detect whether KAP7.1 and KAP8.2 gene were expressed in heart, liver, spleen, lung and kidney tissues; and in situ hybridization to detect KAP7.1 and KAP8.2 gene expression location.(1) qRT-PCR result showed that the expression of KAP7.1 and KAP8.2 gene in the secondary hair follicles are significantly higher than that in the primary follicles, relative quantitative analysis obtained that KAP7.1 was 2.28 times, while KAP8.2 was 2.71 times. Differential expression of these two genes demonstrated that KAP7.1 and KAP8.2 gene may be the main reason with different qualitied cashmere, both KAP7.1 and KAP8.2 gene were high expression in the secondary follicles. Therefore, we speculated that they had an important role in regulating cashmere fineness.(2) Semi-quantitative RT-PCR results showed that KAP7.1 and KAP8.2 mRNA was not detected in the heart, liver, spleen, lung and kidney tissues, explain that KAP7.1 and KAP8.2 genes were specifically expressed in hair follicles, participating in hair formation. (3) ISH analysis showed that KAP7.1 and KAP8.2 gene have a strong expression in the cortical layer and inner root sheath of the primary and secondry hair follicles, indicating that KAP7.1 and KAP8.2 is one the structural proteins of cortical and inner root sheath, involving the formation of cashmere and wool, have an important impact.on physical and chemical properties of hair. In addition, we also found that KAP7.1 gene has a weak expression signal in the hair matrix of secondary hair follicles, demonstrate that KAP7.1 is the one composition of hair matrix for secondry hair follicles, their different expressed locations in Liaoning New-breeding of cashmere goat hair follicle may be one of the reasons for different quality with cashmere and wool.The anagen of hair follicle was chosed in this study, the period, KAP gene expression is the most strongest, which facilitate to depthly know the dynamic changes of KAP genes in hair formation, lay the foundation for understanding KAP genes is how to affect the quality of wool. |
