Avian coccidiosis is due to a protozoan intracellular parasite belonging to the genus Eimeria which multiplies in the intestine of the host, resulting in great economic losses to the poultry industry. Eimeria tenella is one of the most virulent Eimeria species in chiken. During the period of infection, protozoon repeatedly at the intestinal mucosa tissue, proliferation and capillary microvascular were cut, cause massive hemorrhage at a short time, become the leading cause of death. According to statistics, each year billions of dollars was lossed in the wordwild because of the coccidiosis, cost as much as hundreds of millions of dollars in drug control. Currently, chemotherapy is used extensively to control coccidiosis, the resistence of anti-coccidial was more and more serious, due to the long-term use. Traditional Chinese medicine (TCM) have low side effects, safe, and not easy on coccidiosis resistant, long-term use of non-accumulation, TCM is not only kill coccidia, but also enhance the body's immune function of chickens, promote growth of chicken development, improve weight gain and yielding performance.Estabelish the coccidian ifection model, survival rate, relative growth rate, Oocyst per gram (OPG), lesion scores and anticoccidial index (ACI) as the evaluation standard to screening the extaction technology. Extracts wre acquired through ultrasonic, percolation and decocition. Ultrasonic and percolation were passed L9(34) orthogonal design. Through the pharmacodynamic test to determinine the compound in the proportion of each herb, study on the effect between total and separate decoction and the extraction concentration of the TCM. The effective fraction was separated by D101 macroporous resins, elution solvent is distilled water, ethanol, ethyl acetate. Extracts wre confirmed by centrifugation, concentrated and vacuum dried, the yield was calculated. Drinking water administration. The compunts initially isolated three parts, each part give from high, medium and low dose to efficacy trails, choosing the best location and sosage.Then, add some decoquinate in the best fraction as the anti-coccidial, add some Pyrazine sulfonamide sodium chloride in the best fraction as Coccidiosis treatment medications test the effectiveness of prophylaxis and treatment coccidiosis.The results showed that pulsatilla extracts can inhibit the oocyst spores. After 96h cultivation, the average rate of spores more than 80%, extremely individual extract reached 90%, the control group was higher than 90%. Pullsatilla's anticoccidial index (ACI) of the maximum is 156.3, close to the midum efficiency. Can improve the survival rate and reduce the OPG, weight gain significantly. The best extraction technology is enght times the mount of 50% ethanol, ultrasonic 100min; Acalypha and Pteris decocition is well, but when the three TCM decocited together, the ACI will lower than not. The best extraction technology of Acalypha and Pteris is ten times the mount of 50% ethanol, Pecolation extract, the velocity of flow is 7-10ml/min*kg; Extract concentration of compound administered 20ml/L, the ACI is 166.1, while administere 30 ml/L the ACI is 157.8; the active sites of the highest part of the Compound is the fraction of alcohol when isolated by macroporous resin, ACI can reach 163.9. The fraction of alcohol adding Decoquinate for efficient prevention of Coccidiosis, ACI is 181.4, but Add Pyrazine sulfonamide sodium chloride for treatment has little effect,ACI is only 146.8.In summary, this paper identified the compound of the extraction technology, anti-active fraction was enriched after further separated; TCM adding chimecal drug has a strong anticoccidial activity. The water agent easy to use, anticoccidial effect stable and effective.
