The major morphological characteristics of some grape variaties in AkeSu were studied firstly in this thesis. Then, 9 kinds of grape were studied by the method of RAPD molecular marker, which was based on the establishment and optimization of grape's RAPD reaction system and provided the basis for the identification of grape's germplasm resources and the analysis of cultivars relationship on the molecular level.The main results were as follows:The analytical results of morphological characteristics were showed that the structure numbers of leaves were stable. It could reflect characteristics of various leaves. For example, the descibing characteristic numbers of Hong Mu Na Ge and Bai Mu Na Ge were 146-1-192 on everage(ABC-r-S'S method of Galet method). They were different departments of the same variety, had no difference in characteristics of leaves and accorded with the fact. The gathering pictures of eight varieties were drawed by virtue of the various characteristic structure values of leaves and the various characteristic indexes of fruits. The gathering results could get Hong Mu Na Ge and Bai Mu Na Ge together, so the Ma Nai Zi and Wen Su Hong. It could illustrate that the gathering in varieties and be different with the fact. The identification needed other methods, for example, grape varieties above were classified correctly by RAPD amplification technologies.The methods of improved CTAB,SDS and high salt were used to extrat total DNA from grapes.They were detected by UV, electrophoresis and RAPD. The results were showed that the extraction steps of improved CTAB method were simple, It was the best extraction method of total DNA from grapes in Xinjiang. At the same time, the optimal RAPD-PCR reaction conditions had been established as follows: in a volume of 20μl containing template DNA 100 ng, 10×PCR buffer 2.0μl, MgCl2 2.0 mmol/L, dNTPs 0.1 mmol/L, Taq DNA polymerase 1.5 U, primer 0.3μmol/L.Similar modulus between 0.55~0.85 were calculated by NTSYS soft. 9 test materials could be divided into four kinds ingenetic at the distance 0.73 by UPGMA method cluster analysis and analyze various inter-relationships and lay the foundation for classification using the RAPD member mark to the grape resources research.Morphology and RAPD were used for the test method of grape varieties (lines) for cluster analysis. The results showed that the measured morphological characteristics of indicators and cluster analysis had much correlation. Different grape varieties (lines) could be separated by the method of RAPD molecular markers.
