Hypsizigus marmoreus is a kind of edible fungi with good health care function. The research of biological characteristics and cultivation conditions were carried out from 1980s in China. Hypsizygus mushroom nutrient-rich,including the 8 essential amino acids,including lysine,arginine content is higher than normal mushrooms, which contains many important biological active substances,such as Hypsizygus mushroom polysaccharides,anti-fungal protein,glial-binding protein,ergosterol and Hypsizygus mushroom polysaccharide has a good anti-tumor activity.Based on the true function and good health Pleurotus cornucopiae broad market prospects,this paper shake flask method Hypsizygus mushrooms in submerged culture conditions,namely,speed,temperature,pH,inoculum size,bottle volume, incubation time were studied,which contain rotational speed,temperature,pH, inoculum size,bottle volume and incubation time. The results showed that the optimum submerged culture conditions for Hypsizigus marmoreus were listed as follows:the stirring speed at 120 r/ min,temperature at 25℃,pH at 4.0-7.0(not necessary to regulate),inoculation volume at about 10 mL (250 mL erlenmeyer flask, aeration ratio at 100 mL ),the aeration ratio at 100 mL(250 mL erlenmeyer flask),and the culture period on the 7th day. At the same time through experimental verification of the findings are credible.View of the current Hypsizygus mushroom polysaccharide extracts commonly used hot water extraction,solvent extraction and enzymatic extraction of the status quo this paper hot water extraction for innovative improvements. Although the reference to the yield of thermal flooding lower than the enzyme,but the process is simple,easy to operate and economical. Therefore,this formulation to extract the thermal flooding Hypsizygus mushroom polysaccharides. Extraction conditions including extraction temperature,the materia1-water ratio,extraction time,ratio of ethanol to extract and extraction times on the yield of polysaccharides were investigated by the single factor experiments. On the base,we do the orthogonal test to optimization of extraction conditions for Hypsizigus marmoreus polysaThrough the orthogonal test,the optimum technology conditions for extracting polysaccharide from Hypsizigus Marmoreus were obtained as follows:the materia1-water ratio of l:30,extracting for 3h at 90℃,and extracting twice.Antiblastic Activitiy of Hypsizigus marmoreus mycelium and fruit-body Polysaccharides. These crude extracts were tested for antimicrobial activitiy to Bacillus subtilis,Escherichia coli,Staphylococcus. Aureus,Saccharomyces,Aspergillus niger and Rhizopus sp with the agar diffusion method. The results showed that the polysaccharides liquid had an inhibiting effect on Bacillus subtilis,Escherichia coli, Staphylococcus. Aureus,Saccharomyces. The MIC was in the rang of 10-5mg/mL. Hypsizigus marmoreus mycelium polysaccharides and fruit-body polysaccharides had no effect on Rhizopus sp. and Aspergillus niger.Although the Hypsizygus mushrooms have better nutritional and medicinal value,but the depth of its development is still limited,but Hypsizygus mushroom polysaccharides rates were generally not high , so Hypsizygus extraction of polysaccharides from mushrooms there are still a lot of room for improvement,such as the walk in front of more than lentinan extraction methods and high extraction rate can be considered applicable to Hypsizygus mushroom polysaccharides to improve the extraction rate of polysaccharides. |
