Abstract/Summary:
Houttuynia cordata Thunb.,a kind of perennial herb with edible sprout,spear and rhizome,is a important traditonal drug plant with many properties such as anti-inflammatory and antibacterial effects,antivirus function and immunological enhancement.Now,it is supposed to be the very potential plant resource for food and drug by the Ministry of Health.This study in one hand searched for the best cultural condition for the inducement of callus and proliferation through the dynamic analysis of inducement and proliferation of callus and growth cycle to provide the necessary condition for the available medicinal constituent from cell culture.In the other hand,tried find out the most effective antibacterial population by the study of different kinds to offer the theoretical basis of genetic resource protection,Selecting and utilization of Fine Varieties.The chief results were as follows:1.1/2MS+ 6-BA 0.5mg/L+2,4-D 1.0mg/L+2~3%sugar,PH6.0,24h darkness culture were the best conditions for the induction of callus according to a series of single factor experiments.Paraffin sections showed the most number of elementary cells helpful for the subculture in the microscope.2.MS(0.8%agar+3%sugar)+ 2,4-D 0.2mg/ L+6-BA0.5 mg/ L was suitable for proliferation and browning control.VC,PVP were active on controlling explant browning, and pvp was the best.2g/Lpvp made the lowest effects on inhitbitting browing.Different light application time made different.In the early time of cultivation,darkness was helpful for inhibitting browing.In normal condition,shorten the time of subculture could effectively reduce the poisonous action of quinone.3.Loose callus in pale yellow flourished and the increase became the highest when inoculum concentration reached 3.0g/bottle.The green-weight appeared as "s" in the growth process and the life circle was 30d.The growth curve could be divided into 3 periods:delayed phase(0~8d),exponential phase(8~16d),stationary phase(16~240d).In the exponential phase(8~16d) cellular metabolism blooms and callus grew quickly reaching the top,8.47g/bottle on the 16th,then went into the stationary phase with lower metabolism and not obvious weight change.4.The best condition for the extraction of essential oil by using orthogonal test was:2 hours' immergence,10 times of water,7 hours'extraction. 5.Antibacterial experiment showed that essential oil was of no use for blue mold and Aspergillus niger,but was of great effect on Staphylococcus aureus and had certain function on Esherichia coli and Bacillus subtilis.8%of resource oil's effect was close to 4×102 U/ml of Benzylpenicillin sodium salt and the more the better.According to the study, population Huoshan population reacted to Staphylococcus aureus better than Chaohu and Guangde,the populaiton in Tongling appeared as the worst;Chaohu,Chuzhou and Huoshan population reacted better to Esherichia coli;Guangde,Huoshan and Chaohu population beeter to Bacillus subtilis,and Guangde appears the best,then Chaohu and Huoshan,Dujianyan showed the worst reaction.6.Anti-inflammatory experiment showed that all:essented oil of all kinds of oil shows different suppression effects to the ear swelling of rats and Chuzhou populatin lagerly differs from the model grooup;All the experimental subjects suppressed feet swelling of rats significantly compared to the model and among them Chaohu,Hushan,Chuzhou and Hefei populaiton at every time differ from the model apparently;All the subjects show suppression to TNF-αin different degrees.Huoshan,Guangde,Shitai,Hefei and Wuhu population showed very differently from the model and Huanshan,Hubei and Chuzhou plants got significant difference;Tongling,Jixi,Huangshan and Hubei population show suppression to IL-1 significant difference from the model;All the subjects show significant suppression to the proliferation of granulation of rats,and Chaohu,Jixi,Huanshan and Huoshan plants appeared significant difference from the model and Sichuan,Hubei,Guangde,Shitai,Chuzhou and Hefei population had very significant difference from the model.
