Browning takes place generally in tissue culture of peony,so there isn't many young plants can be used in production and scientific research.This paper do some systematic research on browning.This paper used Heilongjiang Wild peony and cultivation peony as experiment materials, do some systematic research of the explant browning after inoculation in the process of callus induction,with a view to establish a renewable peony optimization technology system.Experiments around the browning in the process of tissue culture-induced, multiplication and differentiation conducted a study on disinfection,exogenous growth regulators,endogenous growth regulators,inhibiting browning methods,callus maintaining culture,and also compared the correlation about browning of explants derived different parts different growth periods,different environmental conditions.Then get an effective way to inhibit browning through the systematic research of inoculation,callus multiplication and differentiation.The result show that:1.Explants from the early stage of culture,add absorbent PVP1g/L in the medium, effectively control the two types resources test peony leaves,leafstalk,stem and petals explant browning,in the 30d,its inhibit browning rate was 100%.2.Explants from stage inoculation.Add the suppression reagent,use the suppression reagent pre-treatment before inoculation,change the conditions of culture have obtainned an effective inhibitory effect.Among them,add anti-oxidants citric acid,Victoria C and citric acid pretreatment,low temperature culture,dark culture can effectively alleviate browning.Citric acid on inhibition of the browning petals is the most effective way,in the 30d,only can see the medium of explants 0.3cm radius around light brown,no browning explants itself.Two Peony of the browning rate lower than that of the control respectively 84.38%and 90.63%;vitamin C pretreatment of wild species inhibit browning,leaves best browning rate fell to 25%.Citric acid pretreatment inhibit browning the resources on the cultivation stem,the browning rate of 37.50%3.In the process of callus multiplication and differentiation,add PVP to good and bad callus, the effect of inhibited browning to good callus is significant,the not browning individual reach to 100%.The effect of callus induction in different developmental stages,based in Harbin in late April to early May,the two peony sources of stem and leaves is the best,in the 14d can get good callus.4.Different basic culture medium and growth regulators have varying degrees influence on browning,and low concentrations of exogenous growth regulators can relieve explant browning effect.Which high concentrations of 2,4-D on the two explants peony facilitate the browning, the browning rates were 87.50%and 66.67%,lower concentration browning rate of 2,4-D is 50%and 39.40%.5.Browning correlated with PPO activity,getting materials in different parts of leaves,petioles,stems and different developmental stages,the PPO activity at different levels between two Paeony.Resources in different developmental stages and parts is an effective way to inhibit browning.Cultivars stem in the time of leaf-expansion period is the most appropriate time and site materials,wild species stem in the time of leaf-expansion flourishing period is the best.
