The 31 protoplasted and 9 sporulated homokaryons were obtained from cultivated strains WX-1 and L205 of Lentinula edodes(Berk.) respectively for cross breeding. After symmetric cross combination 131 hybrids were obtained. Among the hybrids 43 were screened out because of their too slow growth rate and unsuccessful colonization in cultivation log. The cultivation experiment was conducted with remaining 88 hybrids, and each hybrid was replicated 6 times. Though 58 of 88 hybrids produced mushrooms but 20 of them were produced very few and misshapen mushrooms. The agronomic characters of rest 38 hybrids were taken to compare performance with parental strains. Gray correlation degree analysis was applied to analysis 8 agronomic characters. Lastly, 14 excellent hybrids were screened out such as L9-32,L3-21,L1-34 and so on.The results showed that hybrids L3-21,L2-10,L1-34 and L3-34 were excellentover parental strains in respect of comprehensive characters.Amplified fragment length polymorphism (AFLP) was done for analyzing and compared the genomic DNA of hybrids with their parents. Four pairs of primers produced 160 fragments, on an average 40 fragments per primer, 94 were polymorphic (58.75%). Un-weighted pair group method with arithmetic mean (UPGMA) cluster analysis was used and obtained the dendrogram.The main research results obtained are as fallowings:1. The genetic similarity coefficient of hybrids ranged from 0.7791-0.9596. From secondary screened strains are divided in 3 categories at 0.7791 similarity coefficient, Parents wuxiang No.1 and L205 were clustered separately in groups I and II respectively, most of hybrids were clustered in two groups. Hybrid L2-10 was clustered separately in group III.2. Protoplasted monokaryons hybrids clustered in group II, the genetic similarity coefficients among L69-15, L91-15 and L91-7 were high. Among them, the genetic similarity coefficient between L91-15 and L91-7 was highest, it was 0.9609. The genetic similarity coefficient between L69-15 and L91-7 was 0.9271.3. The equal weight correlation degree of strains L3-21, L205, L1-34, L91-15, L91-7 were 1, 2, 3, 4, 6 respectively, they had a higher correlation coefficient with ideal strain. And the five strains were clustered in group II. These results showed that, the genetic background and agronomic characters of hybrids had a definite relationship. But it was not in a simple linear relationship, and needed large number of demonstration.The experimental results indicate that, the molecule markers of genetic polymorphism not only revealed the genetic diversity and relationship of the hybrids of Lentinula edodes, but also provided a new idea of the way of genetic polymorphic molecule markers guided conventional cross-breeding.
