The Effect Of Sodium Pyrosulfite On Callus And Cells Suspention Culture In Tomato

In this experiment, the effects of Sodium pyrosulfite (SMB) on the callus and cells suspention culture of vegetable were discussed by combining suspention cells and SMB. The results supply some theoretics gist for the future in-depth research. The factors,which influenced the maintainment and regeneration of suspension cells lines. Such as sugar, medium,grows regulator. The main results were as follows:(1)The perfectly antisepsis measures medium which sterilize seed of cultivating antisepsis seeding was liquor of 10%NaClO and a drop of tween80,the time of disinfection is 30 minutes.Another mesure was liquor of 10%NaClO and 0.1%HgCl₂,the respective action time was 20 minutes and 3 minutes.(2)The pH of medium were changed by appending SMB,so the results were reducing seminal germination capacity and bad effect of offspring growth.(3)Callus were advanced form when their were cultured on MS medium with 0.2 mg/L 2, 4-D,0.2 mg/LNAA and 0.05 mg/L ZT.The best concentration sugar is 3%,which adapt to callus growth.(4)The best concentration combination was 0.2 mg/L 2, 4-D,0.2 mg/L NAA and 0.05 mg/L ZT,which were good for the suspention cells culture.The best sugar concentration was 3%.(5)The approximately effect trend of SMB on suspention cells culture were descendent.The trend contents include solute protein content,MDA content and POD activities.Their contents were differ from natural growth condition.
