Differential Proteomics On Dwarfing Mechanism Of Peach (Prunus Persica) 'Yu Dwarf Rootstock No.1'

Abstract/Summary:
In this study the leaves from Yu Dwarf rootstock No.1 and Kansu peach in different growth periods of April, June, August and September were selected as material, and differential proteomics were utilized to study dwarfing mechanism of Yu Dwarf root- stock No.1. The main results are as follows:1. An optimized 2-DE system was used to establish 2-DE technology suitable for leaf proteins of peach (Prunus persica). By compared effects of two different lysis buffers, improved 2-DE parameter, and selected appropriate loaded sample, 2-DE gels were successfully obtained. The improved 2-DE system resulted in a 2-D protein profile of higher resolution and higher protein yields as analyzed by Image Master 2D software, indicating that it is a suitable 2-DE system for analyzing leaf proteins of peach.2. By using the improved 2-DE system to isolate leaf proteins of Yu Dwarf rootstock No.1 and Kansu peach, 2-DE gels of four different growth periods were acquired, and then analyzed by Image Master 2D software. The result showed that:①On Yu Dwarf rootstock No.1 gels of April, June, August and September, 781, 638, 621, 516 proteins were respectively detected. And 763, 640, 618, 524 proteins on Kansu peach gels of April, June, August and September, respectively. Most of proteins were distributed in the range of Mr 12~72KDa and pI 4~7.②According to Scatters plot of proteins, the proteins correlation coefficient of Yu Dwarf rootstock No.1 and Kansu peach were higher in August and September, but lower in April and June.③On gels of Yu Dwarf rootstock No.1 and Kansu peach, twenty-two differential proteins were detached, of which fifteen proteins present only on gels of Yu Dwarf rootstock No.1, four proteins exist only on gels of Kansu peach, the other three proteins exist both on gels of Yu Dwarf rootstock No.1 and Kansu peach, but differ in abundance or appear in different periods.3. Twenty protein spots excised from 2-DE gels were destained, reduced, alkylated and then digested with trypsin. Through MALDI-TOF/TOF-MS analyses on ABI 4700 Prote- omics Analyzer, both MS and MS/MS data were acquired. After database searching in NCBInr, eighteen spots of which were identified as thirteen proteins, they are correspon- ding to At1g17880/F2H1510, high molecular weight heat shock protein, ribulose-1,5- bisphosphate carboxylase /oxygenase large subunit, oxygen-evolving enhancer protein 2, eukaryotic initiation factor iso4E, tubulin beta-7 chain, nucleosome assembly protein 1-like protein 3, fructose-bisphosphate aldolase, At3g04620, photosystem II reaction center PsbP family protein, proteasome subunit beta type-6 precursor, glycine dehydr -ogenase [decarboxylating], polyphenol oxidase precursor. The identified proteins are involved in the processes of photosynthesis, response to stress, energy and amino acid metabolism, cell cycle regulation and growth regulation.The roles of these proteins in dwarfing mechanism of'Yu Dwarf rootstock No.1'as well as future work were dicussed.
