Avian infectious coryza is an acute respiratory disease caused by Avibacterium paragallinarum, which can mainly inhibit chiken growth and reduce egg production. Coryza occurs worldwide, and it can be cured by drugs, following wih great economic losses since it is easy to relapse. The membrane proteins with immunogenicity perform various physiological functions in cell regulation, responsible for transformating substances and transducting signals between cell and environment. Most of the diseases were caused by the deficiency of a particular membrane protein, the number of biological research against activating or inactivating the membrane protein has been increasing more and more, but seldom reported on membrane protein of Avibacterium paragallinarum.In order to further optimize the prevention measures of infectious coryza and investigate the immunogenicity of membrane protein of Avibacterium paragallinarum, the growth changes of three reference strains: Avibacterium paragallinarum A (221), B(0222) and C (Modesto) was observed and compared using TSA and TAB medium under the same culture conditions in this study, to determine the best culturing time; the membrane protein of three strains-Avibacterium paragallinarum A (0083), B(0222) and C (Modesto) were extracted by FOCUSTM Global Fractionation kit and examined through sodium dodecyl sulphate-polyacrylamide gel electrophoresis(SDS-PAGE) and two-dimensional (2-D)electrophoresis (pH4-7) to study on the preparation method and the difference of membrane protein of Avibacterium paragallinarum, experimental results show that:1、Under the same culture conditions, C-type have had the shortest survival time, and while A, B-type survived longer, and the growth plateau of the3-three type strains occurred at in training to13h-14h.2、Three different serotypes of Avibacterium paragallinarum presented similar SDS-PAGE patterns, with seven main strips of the same molecular weight in each samples, The percentage of serotype A, B and C in the total protein weight is respectively74.06%,75.03%and67.56%. In addition, the molecular weight of three bands were somewhat identical, whereas the content of them were significantly different.3、There were21disparate points of membrane protein was screened from three types2-D electrophoresis maps of membrane proteins. Seventeen valid membrane points were identified as nine proteins combined with Tandem Mass Spectrometry and gene database. The biology information displayed that the proteins performed various physiological functions such as bacterial immunity、 participating in regulating cell metabolism and defensing harmful substances.It has laid a good experimental basis for developing new vaccines, revealing the immune mechanism and pathogenic mechanismthe of Avibacterium paragallinarum.
