FMDV is the causative agent of FMD,a highly contagious disease of cloven-hoofed animals. Owing to FMDV of variation was vulnerable which usually bring differences of immune and epidemic strain that includes virulence,prevalence states,antigenicity,so the prevention of the current FMDV was extremely important.VP1 gene of O type FMDV and PET-28a and PET-41a prokaryotic expression vector linking then through time,temperature,IPTG concentration to optimize the expression conditions,meanwhile application ethanol and dithiothreitol to promote the expression of soluble protein,but the result Unsatisfactory.The analysis of its particle crystal in 1989 provides a possible way to study its characteristics of structure and function,not only beneficial to the prevention and treatment of FMDV,but also reveal catalytic point of enzyme, protein form(such as the formation of inclusion bodies),the combined and identify of between the material from the composition and conformation of protein,so that help our comprehensive understanding structure,function,polymorphism of protein deeply.Through the use of a variety of software and SWISS-MODEL service platform to predict the secondary structure of O type FMDV,simultaneously compared VP1 protein and 1fod1 from PDB database found raise a helix 5-7 residues and lost fourβ-tune that located at 4-7,5-8,102-105,135-138 residues,other secondary structure had not changed.The result point to amino acid changes can change the secondary structure of VP1 protein,led to formation of new secondary structure,but some amino acid changes without affecting the secondary structure of pre-existing protein,which provides a new perspective for conservative and the process of biological evolution.The use of SWISS-MODEL service platform,we predict the three-dimensional structure of VP1 protein of O type FMDV and assessment space structure.Through the similarity comparison of protein three-dimensional structure found some changes of amino acid lead to changes of space Structural that for antigen site of screening,differences in virulence,prevalence of viuse, interaction between the protein provides a physical reference.
