Elite variety is the base of high yield,and impure variety will lower yield.So,it is good for seed standardization,variety approval and indentification,property protection,to indentify variety and analyze variety purity.Genetic diversity and relationships of germplasm resource in crop is the base of breeding. Evaluation of genetic diversity among cultivars can help to assign parent and predict genetic variety and heterosis,which it is the key of implementing breeding object.In this paper,Based on the SSR analysis,we analyzed the genetic diversity and constructed the DNA fingerprinting of 89 varieties from Jilin province.The main results were as follows:1.Determine the optimum response to the SSR system:the reaction system 25μl,including the template DNA 40 ng,primers 0.5umol,dNTPs 0.4mmol,10×buffer 2.5μl,TaqDNA polymerase 1.5u,to fill remaining ddH2O.Amplified procedures for:94℃predegenerated 5 min;94℃degeneration 45 s,55℃rehabilitation of 45s,72℃extension of 1 min,40 cycles;last extension temperature of 72℃10min.2.Polymorphism through 20 pairs of primers the differential results,primers satt586 and primer satt530 the most efficient identification,to identify five varieties;satt002 able to identify four species;sct188 able to identify three varieties.Primer satt586,satt530,sct188,such as the Shannon in the value of more than 1.35,in the identification and purity of soybean varieties can be identified priority.3.There are 110 alleles were amplified by 20 SSR primers and with the mean of 5.5,satt586 and satt329 respectively amplified 10 alleles,satt022 amplified 3 alleles;Each alleles of the SSR Shannon index for the distribution of 0.7068-2.1225,with a mean of 1.3193,the index is the largest primer satt586,followed by satt239.4.Genetic varieties for similar coefficient of the range of 0.385-0.936 changes,the overall average of 672,similar to larger coefficient,small genetic differences between the species.Between the various groups to in a variety of Jiyu(group 1),and Jinong(group 2) hereditary genetic distance(0.2312) recently. Jiyu(group 1) with jiunong(group 4) genetic distance(0.6216) farthest.5.Based on the results of the SSR markers,for the varieties of cluster analysis for varieties divided into three groups and a special varieties.Referred to as SGs(SSR-based Groups):SGⅠincluding 71 varieties, SGⅡincluding nine varieties,SGⅢincluding eight varieties,special varieties Bainong 9.
